Objective:To screen anticancer extractions of Chinese herbs Zanthoxylum nitidum(Roxb.)DC.& Plumbago zeylanica L and study their molecular mechanisms in vitro.Methods.1.Experiment on the extractions of Chinese herbs Zanthoxylum nitidum(Roxb.)DC.& Plumbago zeylanica L and tumor cell lines SPC-A-1,7111, Ecv2,Tca8113 in vitro,the growth inhibitory effects was evaluated by MTT testing.2.Experiment on Nitidine Chloride & AgLA2 and tumor cell lines SPC-A-1,7111,apoptosis-inducing effects was investigated by DNA agarose gel electrophoresis,cell morphology and Elisa.3.Experiment on Nitidine Chloride & AgLA2 and tumor cell lines SPC-A-1,7111,RT-PCR and Real-time PCR was used to measure the expression of bcl-2,bax and C-erbB-2 mRNA,immunocytochemistry was used to measure the expression of bcl-2,bax and C-erbB-2 protein,Flow Cytometry was performed to measure the concentrations of NO in cells.Results:1.After MTT testing,several anticancer extracts were screened out,which were Nitidine Chloride,(PtLA)â… ,(PtLA)â…¡,(AuLA)â… ,(AuLA)â…¡, AgLA2,ZnLA2,SnLA,Mo2LA2,plumbagin,CuPL2,NiPL2,CoPL2.2.Treated with Nitidine Chloride & AgLA2,SPC-A-1 and 7111 cells underwent apoptosis. Nuclear DNA of apoptosis cells displayed ladder bands characteristic of internucleosomal DNA fragmentation.Apoptosis cells demonstrated compaction and segregation of the nuclear chromatin,condensation and vacuolus of the cytoplasm.The average concentrations of Caspase-3 in the drug-treated groups was higher than the control groups.3.Treated with Nitidine Chloride & AgLA2, bcl-2,C-erbB-2 mRNA and its protein expression decreased while bax mRNA and its protein expression increased,the concentrations of NO in the drug-treated cells upgraded too.Conclusions:Nitidine Chloride and AgLA2 can induce SPC-A-1,7111 cells to apoptosis.The first point of the mechanism may be changing the ratio of bax/bcl-2 and the set-point of apoptosis,and it makes the apoptosis potency take the dominant position in the tumor cells.The sencond point may be inhibite the activation of the protooncogene C-erbB-2 and the third point may be upgrading the concentrations of NO in the cells to enhance the antitumor effect.
