Objective:To develop an HPLC method for simultaneous determination of5active compounds. Thus, in order to probe the possibilities and experimental data for the replacement, the difference of content and chemical composition between Zanthoxylum nitidum (Roxb.) DC. and Zanthozylum nitidum (Roxb) DC.f. fastuosum How ex Huang is performed by the content determination and fingerprint analysis.Methods:1. The initial evaluation sources preparatory evaluation of the commercial materials of Zanthoxylum nitidum (Roxb.) DC. was performed by morphological study and pharmacognosy research.2. The determination and comparison of the5content in both medicinal materials, magnoflorine, hesperidin, nitidine chloride, ethoxychelerythrine and toddaloactone, were carried out by HPLC.3. Chromatographic fingerprints analysis of each batch of commercial medicinal materials of Zanthoxylum nitidum (Roxb.) DC. and Zanthozylum nitidum (Roxb) DC.f.f.was established by HPLC, and the shared and different constitutions was compared by3kinds of statistic methods, system clustering analysis, angle cosine and similarity computations.4. The comparison of the components and the differences of contents between Zanthoxylum nitidum (Roxb.) DC. and Zanthozylum nitidum (Roxb) DC.f.f. in the the decoction products were finished as mentioned above.Result:1. Except for a few batches of the medicinal material, most of the batches are Zanthozylum nitidum (Roxb) DC.f.f.2. The assay studies show that the major constituents in the Zanthoxylum nitidum (Roxb.) DC. are ethoxychelerythrine and nitidine chloride, few batches contains hesperidin. While magnoflorine and toddaloactone were the main constitutions in Zanthozylum nitidum (Roxb) DC.f.f., and toddaloactone was unique in it. And5components in the root were significantly higher than its stems.3. Fingerprint data shows that a huge difference in chemical compositions between the medicinal materials. The former mainly contained alkaloids; while the latter fingerprint appeared a lot of ultraviolet absorption peaks which were similar with Toddalolactone, and the peak areas of each batch varies from each other, and it reflects that the content varies with its production place and its culling seasons.4. Decocting results suggest that the contents of the5chemical components in water decoction were significantly lower than the contents in the medicinal materials; The content of Toddalolactone and Hesperidin in Zanthozylum nitidum (Roxb) DC.f.f. decocting were relatively high.Conclusion:The present experiments show that most of Zanthoxylum nitidum (Roxb.) DC. selled on the market actually are Zanthozyhim mtidum (Roxb) DC.f.f., and the compositions and contents in it are highly different from Zanthoxylum nitidum. Different compounds have corresponding pharmacological activities, whether the difference of chemical composition and content predict the possibility of the replacement of Zanthoxylum mtidum (Roxb.) DC. depends on further pharmacology, pharmacokinetic and toxicologic research.
