The Xiaoaiping tables produced from the raw material of Tongguangteng [the dry stems of Marsdenia tenacissima Robx. Wight et Arn, belonging to Marsdenia R. Br. of Asclepiadaceaes] have an excact curative effect on clinical curing of cancer. But the internal or abroad studies and reports about the components in Tongguangteng are lacking and superficial, just point out that it contains many components such as saponins, alkaloids, organic acids, polysaccharides, colophonies, pigments and so on, and there's no study or report about it's active anticancer components. Tongguangteng has complicated origins as well as many synonyms.In our country, chlorogenic acid is taken as the index components for content determination of the injector produced from the raw material Tongguangteng. This evidently isn't enough to ensure the security, availability and quality stabilization in clinical using. In this article, we do some thorough research on the physical base and react mechanism of it's antitumour activity to develop a safe, effective and quality controllable modern traditional Chinese medicine,this article.1. We have completed the textual study on Tongguangteng, summarized its synonyms as well as clarified the isonyms by looking up many literatures and ancient Chinese medical books. Tongguangteng originates from Yunnan is the same crude drug as Naijiangteng originates from Chengdu, they are synonyms, while Wuguteng, another name of Tongguangteng, and Wuguteng originates from Guizhou are homonyms. We for the first time report that there's the herbal Marsdenia tenacissima distributes in Sichuan province. The pharmacognostical identification of it has been carried out by the botanical and microscopic characters. 50 % and 70 % EtOH elutions were determined as active fractions by MTT text in vitro, but only 70 % EtOH elution showed positive Lieberman-Burchard reaction, indicating that it contained steroidal glycosides. Thus, we mainly investigated the 70 % EtOH elution and resulted in the isolation of seven compounds: tenacissoside L (Ⅰ), tenacissoside M (Ⅱ), tenacissoside N (Ⅲ), tenacissoside O (Ⅳ), tenacissoside P (Ⅴ), daucosterol (Ⅵ) andβ-sitosterol (Ⅶ). Among them,Ⅰ~Ⅳare new C steroidal glycosides, whileⅤis first isolated from this plant.2. Through MTT experiments, tenacissosides L~P could inhibit the proliferation of tumor cell lines in vitro, such as SGC-7901 Human Gastric Carcinoma cell line, Bel-7404 Human Hepatic Carcinoma cell line, C-26 Colon Carcinoma cell line and Hepal-6 Mouse Hepatic Carcinoma cell line and demonstrate different sensitivity. At the same time, all these tenacissosides have little effect on 293 Human Embryo Kidney cell line and L02Human Hepatic cell line。. Using flow cytometry, we observed that 70 % EtOH elution could induce the apoptosis of SGC-7901 Human Gastric Carcinoma cell line, the cells mostly stay at the period G1, the peak of Sub-G1 can be detected, and it accounts for about 40.58 %.3. Studied the extraction and isolation technology of the raw material Tongguangteng origins from Sichuan, decocted it by water, and used macroporous resin to adsorb it twice, then use macroporous ion exchange resin to purify it。The yield of the saponin gained is about 1%, and the content is more than 90%. Taking the saponin we gained as raw material, carry out the technics of lyopyilization for producing freeze drying injectable powder, and have trial-produced injectable powder of tenacissosides. Use tenacissoside L,tenacissoside M,tenacissoside N,and tenacissoside O as the reference substance to establish a method for Qualitative and Quantificational Analysis, and provide important informations for constituting the quality standard.
