| It is one of important tasks that the science of pharmaceutical analysis should solve the actual problems emerged in the scientific research, development and production of traditional Chinese medicines (TCM). Instructed by the theory of TCM modernization, combined newest achievements in the analytical technology of modern science are applied to the pre-phase research of Tongguangteng (Caulis Marsdeniae tenacissimae, Tenacious condorvine Stem), an anti-tumor TCM with reliable effect and unknown effective constituents.Tongguangteng was first recorded as drug in "Materia Medica of Southern YUN NAN"(Dian Nan Ben Cao) based on herbal textual criteria. Its original plant is Marsdenia tenacissima, belonging to Mardenia R. Br. of Asclepiadaceaes. Its nature and flavor is bitter and sub-cold. It can clear away the heat-evil, expel superficial evil, eliminate inflammation, clear away heat-toxin, relieve cough and asthma and relieve pain by removing obstruction. Its indication is bronchitis, bronchial asthma, cancer, neoplasia and long-standing toxin. More than 10 herbals of different species are currently used as Tongguangteng.Preliminary screening of bioactive compounds, especially anti-mitotic and anti-fungal substances from active fraction by MTT and Pyricularia oryzae bioassay methods, 23 compounds were first isolated and identified with GC-MS method from the ether fraction, i.e. c-sitosterol (1), phthalic acid (2), trans-?(sup9)-octadecenoic acid(3), urs-12-en-3á-ol, acetate (4), silane, cyclohexyldimethoxymethyl- (5), 9,19-cyclolanost-24-en-3-ol,acetate(6), stearic acid(7), á-amyrenyl(8), germanicol(9), lup-20(29)-en-3á-ol, acetate(10), palmitic acid (11), vanillin(12), cis-9,cis-12-octadecadienoic acid(13), oxiraneundecanoic acid,3-pentyl,methyl ester,cis-(14), squalene(15), cholest-5-ene-16,22-dione,3á,26-dihydroxy-,3-acetate,(20S,25R)- (16), campesterol(17), stigmasterol(18), kauren-18-ol,acetate,4a-(19), neoclovene(20), 13,27-cyclours-11-en-3-ol,acetate(21), 9,19-cyclolanost-25-en-3-ol, 24-methyl-(3á,24S)- (22), 13,27-cyclours-11-en-3ol,acetate(23). Another 9 compounds were first isolated and identified with this method from petroleum ether fraction i.e. oleic acid (24), 5,8,11-heptadecatrienoic acid,methyl ester(25), palmitic acid,ethyl ester(26), hexadecatrienoic acid,methyl ester(27), arachic acid(28), n-docosanoic acid(29), cholesta-4,6-dien-3-ol(30), stigmasterol-3,5-dien(31), cholesta-6,22,24-trien,4,4-dimethyl-(32).16 among 28 compounds, isolated from Tongguangteng bought from Yunnan Province were first identified with modern chromotographic and spectroscopic techniques, i.e. two novelcompounds according to the best of our knowledge: 12-β-O-benzoyl-dihydrosarcostin-3-O-β-D-glucopyranosyl (1→4)-β-D-glucopyranosyl (1→4) -β-D-3-O-methyl-6-deoxy-β-D-allopyranosyl(1→4)-O-β-D-oleandropyranosyl(1→4)-O-β-D-cymaropyranoside, named as tenacissosides J(Sub 9), 12-β-O-benzoyl-dihydrosarcostin-3-O-β-D-glucopyranosyl (1→4) -β-D-3-O-methyl-6-deoxy-β-D-allopyranosyl(1→4)-O-β-D-oleandropyranosyl(1→4)-O-β-D-cymaropyranoside, named as tenacissosides K(Sub 16). 10 compounds are first reported from the species: olean-18-ene-3-acetate(Sub 2), stigmasterol(Sub 3), saccharose(Sub 14), marsdekoiside B(Sub 6), marsdekoiside D(Sub 4), succinic acid(Sub 5), di-pyrazolidin-1-yl-methanone(Sub 8), conduritol(Sub 10), dihydro-conduritol(Sub 12), marsdeoreophiside B(Sub 13). Four compounds have been reported from the species:β-sitosterol(Sub 1), stearic acid(Sub 7), palmitic acid(Sub 11) and α-amyrin acetate(Sub 15). Among them, Sub 1, Sub 3, Sub 7 and Sub 11 have been identified from ether fraction with GC-MS method.Tongguangteng presents in vitro various bioactivities of growth suppression on the human osteosarcoma cell Saos-2, human gastric carcinoma cell SGC-7901 and human hepatocarcinoma cell Bel-7404 and so on.
