Atherosclerotic renal artery stenosis is the main cause of secondary hypertension, therefore more attention was payed to it.Antihypertension drugs is needed by patients with or without the atherosclerotic renal artery revascularization.We assumed that the antihypertenison drugs have therapy effect on the myocardium by decreasing the blood pressure and regulate the myocardium apoptosis genes,so the experiment was designed to confirm these hypothesis.Objective:To evaluate the effect of antihypertension drugs on the blood pressure,we make the Goldblatt rat model by improving the methods.We also compare the index that including the pathology changes of the myocardium,the anti -inflammatory factor in the serum,the inflammatory factor in the serum,mitochondria peroxidation in the myocardium cell,the index of myocardium apoptosis,the apoptosis gene's expression in myocardiac cells and we want to reveal the therapy effect of the antihypertension.Methods:One side renal artery of the rats was narrowed by operations to made the animal models.The antihypertension drugs including Enalapril and Nifedipine was given to the aninmal model.The immunohistochemistrial methods,the pathological techonologies and the molecular biological methods were used to observe the effect of the antihypertension drugs on the blood pressure,the pathological changes, the cardiac function and myocardium mitochondrial peroxide,the serum inflammatory and anti-inflammatory cytokines,the extent of myocardium apoptosis and the expression of the apoptosis gene.Results:One week after the operation,the blood pressure of the rats in model groups is significantly 20%higher than those in the control groups.The blood pressure of the groups treated with antihypertension drugs is lower than that of the model groups(P<0.05).The antihypertension drugs can improve cardiac pathological changes;increase the cardic function significantly,also it can decrease the inflammatory cytokines and increase the anti-inflammatory cytokines in the serum,and increase the activity of ATP enzyme,glutathione peroxidase,SOD and decrease the content of MDA in mitochondrial,reduce the extent of apoptosis and decrease the expression of the apoptosis genes including the Caspase3,NF-κB and increase the expression of the Bcl-2.Conclusions:1.It is more convenient to make the rat model of renal artery stenosis by ligating the left renal artery using silk-thread with a dorsolateral approach.2.The antihypertension therapys has the anti-inflammatory,anti-mitochondrial peroxide effects.3.The antihypertension threapy can improving the myocardiac pathological changes,ameliorate the cardial function by decreasing the blood pressure and decrease the extent of the mycardic cell's apoptosis by regulating the expression of apoptosis genes to repair the cardiac remodeling.
