Objective A rat model of acute lung injury of after orthotopic liver transplantation was made first,and then to explore oxidative/antioxidative and inflammatory reactions associated ICAM-1 and NF-κB in the pathogenesis of liver transplantation-induced lung injury in adult rats;to study effects of green tea polyphenol on liver transplantation-induced lung injury.Methods(1)Making an adult rat model of liver transplantation-induced lung injury:Seventy-two rats-were randomly divided into two groups,the control group(8rats)was given anaesthesia and operation of opening abdomen,and liver transplantation group(64 rats),was given operation of orthotopic liver transplantation.Experience was over in 1h,4h,8h,and 12h after orthotopic liver transplantation respectively.Arterial blood gases were measured,then PaO2/FiO2 was calculated.Lung coefficient,W/D value,WBCs and neutrophils in BALF were counted.The total protein contents in BALF was calculated.TNF-a,IL-1β, and IL-8 levels in BALF and serum,MDA,SOD,and MPO levels in lung and serum were assayed respectively.Lung histopathology was assessed with hematoxylin and eosin stain.NF-κB was assayed with IH.TNF-a and IL-1βin lung was assessed with WB.(2)based on former rat model of liver transplantation-induced lung injury:thirty-two rats were randomly divided into three groups,the control group(8rats)was given anaesthesia and operation of opening abdomen;liver transplantation group(8 rats),was given operation of orthotopic liver transplantation;and green tea polyphenol group(12 rats)was given 100mg/kg green tea polyphenol 1h before orthotopic liver transplantation. Experience was over in 4h after orthotopic liver transplantation.Arterial blood gases were measured,then PaO2/FiO2 was calculated.Lung coefficient,W/D value,WBCs and neutrophils in BALF were counted.The total protein contents in BALF was calculated.TNF-a,IL-1β,and IL-8 levels in BALF and serum,MDA, SOD,and MPO levels in lung and serum were assayed respectively.Lung histopathology was assessed with hematoxylin and eosin stain.ICAM-1 and NF-κB was assayed with WB and IH.Results(1)The PaO2/FiO2 decreased after orthotopic liver transplantation.Lung coefficient,Wet-to-dry weight ratio increased,the WBCs,neutrophils,and protein contents increased.The levels of TNF-a and IL-1βin both BALF and serum increased to maximum at 4h,and the levels of IL-8 in BALF reached maximum at 4h.The MDA and MPO contents in lung and serum increased,SOD activities decreased.NF-κB contents in lung increased.TNF-a and IL-1βcontents in lung increased.(2)Contrast with liver transplantation group,the PaO2/FiO2 value of green tea polyphenol group increased,and lung coefficient,W/D decreased.The WBCs and neutrophils,protein contents in BALF decreased significantly.The levels of TNF-a,IL-1β,and IL-8 in both BALF and serum decreased.The MDA and MPO contents in lung and serum decreased and SOD activities increased. ICAM-1 and NF-κB contents in lung decreased.Conclusions(1)A rat model of acute lung injury of after orthotopic liver transplantation was made successfully.(2)Oxidative/antioxidative reactions, inflammatory reactions,and neutrophils activation in lung are involved in the pathogenesis of liver transplantation-induced lung injury;(3)green tea polyphenol have protective effects on liver transplantation-induced lung injury.
