ObjectiveProbing into the Traditional Chinese Medical Pathologensis of menopausal syndrome.To clarify the function and mechanism of Bushen Shugan prescription on the gene level restraining menopausal syndrome from the reproductive endocrine,expression level of estrogen receptorαmRNA on hypothalamus and pituitary,and reveal the kidney deficiency and liver depression' scientific content of menopausal syndrome.Methods①60 female Wistar rats aged 12 weeks(250g±10g)were divided into normal group,model group,diethylstilbestrol group,Gengniankang group and Bushen Shugan preliminary group(n=12).The later 4 groups of big rats were taken ovariectomized surgery and angered by the end of folder.All rats were irrigated paunches for 4 weeks.All of the experimental animals put to death on the 2th day after the last treatment,and collected serums, plasma and samples,measured and observed the following indexes.②Take the radioimmunochemistry to measure the levels of serum estradiol(E2), testosterone(T)and follicle stimulating hormone(FSH).③Take the radioimmunochemistry to measure the levels of plasma thromboxane B2(TXB2) and 6-prostaglandin F1α(6-keto-PGF1α),then calculate its ratio.④Take the uterus and adrenal gland's wet weight of all rats with electronic balance and convert organs weight coefficient.Observe the changes of uterus and adrenal gland histology under light microscope of each group. Observe the changes of uterus ultrastructure under electon microscope.⑤Take the TUNEL to measure the apoptosis rate of uterus cell in all rats.⑥Take the RT-PCR to measure expression levels of estrogen receptorαmRNA on hypothalamus and pituitary in all rats.Results①The levels of serum E2,T decreased significantly(P<0.01),while the levels of serum FSH increased significantly(P<0.01)in model group. The levels of E2 and T of Bushen Shugan prescription group were improved,but there was no significant difference between Bushen Shugan prescription group and model group(P>0.05).The levels of serum FSH decreased significantly,there was significant difference between Bushen Shugan prescription group and model group(P<0.05).②The levels of plasma TXB2 and its ratio(TXB2/6-Keto-PGF1α)increased significantly (P<0.05)in model rats.Bushen Shugan prescription can restaint the changes.③The uterus and adrenal gland's wet weight decreased significantly(P<0.01)in model rats,Bushen Shugan prescription can restaint the changes.Model rats have obvious anomaly of endometrium and adrenal cortex in histology and ultrastructure,Bushen Shugan prescription can relieve the anomaly.④The apoptosis rate of uterus cell in model rats increased significantly(P<0.01),Bushen Shugan prescription can decreased the apoptosis rate of uterus cell.⑤The expression level of estrogen receptorαmRNA on hypothalamus and pituitary in model rats decreased greatly(P<0.05),Bushen Shugan prescription can increased the expression level.ConclusionThe study has suggested that the kidney deficiency and liver depression is the fundmental pathogenesis of menopausal syndrome. Menopausal syndrome modle rats were firstly taken ovariectomized surgery and angered by the end of folder.By the experimental study,we detected the levels of serum E2,T decreased significantly,but the levels of serum FSR increased significantly,the levels of plasma TXB2 and its ratio(TXB2/6-Keto-PGF1α)increased,the uterus and adrenal gland's wet weight decreased significantly,obvious anomaly in histology and ultrastructure in uterus and adrenal gland,the apoptosis rate of uterus cell increased significantly,the expression level of ERαmRNA on hypothalamus and pituitary decreased greatly in model group.Bushen Shugan prescription can significantly improves the above indexes.According to the experimental observation,it can be conferred that the pharmacological mechanismes of Bushen Shugan prescription in the treatment of menopausal syndrome are through the below aspects:①By plant estrogen which may be included in traditional Chinese medicine to regulate of the hypothalamus-pituitary-gonad axis;②By correction the disorder of the hypothalamus-pituitary-adrenal gland axis to regulate reproductive endocrine;③By raising the expression level of ERαmRNA on hypothalamus and pituitary to enhance biological effect of ER.we deduced that it can be one of the main mechanisms of Bushen Shugan prescription in the treatment of menopausal syndrome.
