Objective: 1. A study of BDNF nanoparticles for lumbar spine nerve root injury. 2. Discuss the pathogenesis and clinical stage of lumbar spine nerve root injury.3. Propose a theory that this injury should be diagnosed earlier and treated immediately; find a high performance new way for clinical treatment. Methods: 1. On-line analytical process lumbar spine nerve root injured patient's clinical data by using data mining technical methods. 2. Analyze morbidity features and surgery outcomes of lumbar spine nerve root injury. 3. Establish lumbar sacrum nerve root compression dog model. 4. Study recovery effect of intrathecally administered biodegradable PLGA-BDNF nanoparticles for the sensory neuron of lumbar sacrum nerve root compression dog model DRG.. 5. Integrating experimental study result with clinical manifestation and therapeutic efficacy, establish the pathogenesis and clinical stage of lumbar spine nerve root injury, propose a theory that this injury should be diagnosed earlier and treated immediately. Results: 1. Decompress the nerve root after lweek,2weeks and 4weeks compression separately, BDNF immunoreactivity of DRG neuron increased, following compression time. 2. There isn't obvious BDNF immunoreactivity difference between 1 week group and control group. The difference is observed at 2 weeks group, 4 weeks group is more obvious than 2 weeks group. 3. Compared with control group, the pathological change is smaller when intrathecally administered BDNF nanoparticles. Conclusions: 1. BDNF expression change of lumbar sacral cord is important for the development of lumbar spine nerve root injury. 2. Therapeutic efficacy of dog nerve root compression is manifest when intrathecally administers BDNF nanoparticles. 3. Define the pathogenesis and clinical stage of lumbar spine nerve root injury, propose a theory that "time and method of gold decompression", find a high performance new way for clinical treatment.
