Neck Orthotopic carotid allograft transplantation was performed by micro surgical techniques. The ex vivo vascular donor was snaked in Adenovirus-mediated human hHSP70 solution. hHSP70 gene can successfully transfect transplantation by snaking method. SOD,NADPH of the transplantation can be effectively actived by hHSP70; hHSP70 decreased the active of MPO, prevented the cells from apoptosis. the ischemia-reperfusion injury was lower. Adenovirus-mediated human hHSP70 transfected the transplantation and then expressed the gene production, the gene production,can decrease the transcription of adhesion molecules (ICAM-1,CAM-1) and reguiation factors(NF-kB ), ihibite the secretion of cell factors (IL-1,IL-6,TNF-a), attenuation the expression of growth factors(PDGF,FGF,IGF-1), the gene production can also increase the apoptosis of the mooth muscle cell. hHSP70 gene expressed gene productiong to protect blood vesseel endothelium cell,inhibite proliferation and move of the mooth muscle cell, relieve the effection of immune and nonimmune factors,it can prevent allograft vasculopathy. Give play to body protection gene that would make this new approach applied in the filed of prevented allograft vasculopathy.
