Effects Of Chongcao Shencha Capsule On Expression Of The ECM And TIMP-1mRNA In Rat Glomerulosclerosis

Objective:To investigate nephroprotective effects and mechanism of CongCaoShenCha capsule in rat forepart glomerulosclerosis induced by adriamy cin.Methods:72 wistar rats were randomly divided into 4 groups:Sham operation group,non-treated nephropathy group,CongCaoShenCha capsule group, Fosinopril Sodium group,glomerulosclerosis rats model were induced by unilateral nephrectomy and injecting adriamycin.The model and normal group were fed with normal food,while the treated group were intraga -stricly fed with CongCaoShenCha capsule(0.47g·kg^-1·d^-1)and Fosinopril Sodium group(7.5mg·kg^-1·d^-1).then the each group was divided randomly into different time points of weeks 6,8,10,and12 to observe urinary protein;BUN,Scr,TP,Alb,Chol,LDL and TG were measured after 8 and12 weeks.All rats were killed in the 12 week, pathological changes were observed by light microscopy and glomerulosclerosis index(GSI)were counted.Immunohistochemistry was used to examine connective tissue growth factor(CTGF),Fibronectin (FN),collagenⅣ(Col-Ⅳ),plasminogen activator inhibitor-1(FAI-1), Matrixmetallopro teinases-2(MMP-2),monocyte chemoattractant protein-1(MCP-1),pla sminogen activator inhibitor-1(PAI-1).The semiquantitative PCR was used to detect the expression of TIMP-1 mRNA.Result:1.Compared with non-treated nephropathy group,urinary protein were decreased at weeks 8,10and12(P＜0.01).2.Compared with non-treated nephropathy group,BUN and Scr of CongCaoShenCha capsule were decreased at weeks 8and12(P＜0.01), while TP,Alb were increased at week 12(P＜0.01). 3.50 percent glomeruli suffering different degree sclerosis in non-tre ated nephropathy group,it show localized glomerulosclerosis,interstitial fibrosis and localized ymphocyte and monocyte soakage.Pathohistologi cal study revealed that CongCaoShenCha capsule group had the least renal lesions in terms of glomerulosclerosis compared with the other groups,and glomerulosclerosis index(GSI)is much higher in non-treated nephropathy group.4.According to immunohistochemical result in CongCaoShenCha capsule group and Fosinopril Sodium group,he expression of CTGF,FN, Col-Ⅳwas Stronger in non-treated nephropathy group(P＜0.05).5.The expression of PAI-1 and MCP-lwas weaker in non-treated nephropathy group(P＜0.05,P＜0.01),but MMP-2 was stronger.6.The expression of TIMP-1mRNA was weaker in Fosinopril Sodium group and CongCaoShenCha capsule-treated group,the later was the weakest(P＜0.05).Conclusion:CongCaoShenCha capsule not only reduced proteinuria,improves renal function,but also degraded the expression of PAI-1,MCP-1 and TIMP-1mRNA in kidney,and inhibited glomerulosclerosis.
