Objective : Cerebral ischemia reperfusion injury is one mainly pathological mechanism of ischemia stroke. The functional disturbance of the network of neuro-endocrine and immune (NEI) can be induced by cerebral ischemia reperfusion. The thesis studies effect mechanism of electroacupuncture acupoints rats with cerebral ischemia reperfusion injury from NEI-regulation angle.Methods: The model of reversible middle cerebral artery occlusion (MCAO) /reperfusion was established by improved suture method in Wistar rats. These rats were randomly divided into 5 groups: normal control group, sham-operation group, model control group, electroacupuncture acupoints group and electroacupuncture non-acupoints group. Furthermore every group was randomly divided into 3 time points-1 day, 3 days and 7 days after reperfusion. We observed the rats neural deficit score, measured serum levels of IL-1β, IL-6 and CORT by ELISA method, tested rats' hypothalamus protein expression ofβ-EP and Glu by immunohistochemistry technique, checked rats' hypothalamic or pituitary mRNA expression of CRF,ACTH,GR and TGF-β1.Results: (1) The rats neural deficit score of electroacupuncture acupoints on group can be improved significantly compared with electroacupuncture non-acupoints group and model control group at 3d,7d of ischemia-reperfusion different points (P<0.05); (2)Serum levels of IL-1β,IL-6 and CORT of electroacupuncture acupoints group can be increased significantly compared with electroacupuncture non-acupoints group and model control group at 1d,3d,7d of ischemia-reperfusion different points (P<0.05, P<0.01) ;(3) Rats' hypothalamus protein expression ofβ-EP and Glu of electroacupuncture acupoints group can be up-regulated significantly compared with electroacupuncture non-acupoints group and model control group at 1d,3d,7d of ischemia-reperfusion different points (P<0.05, P<0.01 ) ;(4) Rats' hypothalamus or pituitary mRNA expression of CRF,ACTH,GR TGF-β1 of electroacupuncture acupoints group can be improved significantly compared with electroacupuncture non-acupoints group and model control group at 1d,3d,7d of ischemia-reperfusion different points (P<0.05, P<0.01).Conclusion : (1) Electroacupuncture acupoints has the obvious auxoaction on neurologic impairment recuperation in cerebral ischemia reperfusion injury MCAO model rat's; (2)Electroacupuncture acupoints can regulate serum levels of IL-1β, IL-6 and promote TGF-β1 mRNA expression which can induce cerebral ischemia reperfusion injury its certain action of brain protection ; (3) Electroacupuncture acupoints can inhibit protein expression ofβ-EP and Glu in rats' hypothalamus, which can induce cerebral ischemia reperfusion injury; (4) Electroacupuncture acupoints can regulate expression CRF,ACTH,GRmRNA and serum levels CORT of HPA axis hormone, which can induce cerebral ischemia reperfusion injury its certain action of brain protection; (5) Electroacupuncture acupoints regulate peculiarly rats cerebral ischemia reperfusion injury. Through regulating immunity cytokine, neurotransmitter and disorder of hormone, Electroacupuncture acupoints can rectify the disorder of neuro-endocrine-immune network so as to produce a protect effect on the brain after cerebral ischemia reperfusion from many target spots and links.
