Objective:This study assessed the feasibility of Xenogeneic ADM as a new soft tissue filling material.Methods:The toxicity of the Xeno-ADM was evaluated in vitro.The Sprague-Dawley rat was used as a model for subcutaneous implantation over a 32 week period.Experiment is divided into four groups:1.Xeno-ADM group with integrity of the basement membrane;2.Xeno-ADM group with removing of the basement membrane;3.Allo-ADM group;4.the control group.Histological dynamic changes of ADM were observed after transplanted 2-32weeks.The levels of expression of IgG,anti-ADM antibody,TNFα,IL-6,IFNγin serum were detected by Elisa,Western and RT-PCR after the Xeno-ADM or Allo-ADM was implanted subcutaneously 2-32weeks.Results:1.It was proved that Xeno-ADM was no significant cytotoxity.2.The basement membrane group was the worst among three groups in biocompatibility during 16 weeks.There was no significance after 16 weeks.3 The highest expression of TNFα,IFNγ,IL-6 in serum was observed when ADM had been transplanted for 8 weeks.The levels of IgG and anti-ADM in serum were highest when ADM had been transplanted for 2 weeks.All the levels tend to be stable and close to normal after 16 weeks.But the level in the basement membrane group was higher than in the other two groups before 16 weeks.Conclusion:1.Xeno-ADM showed no significant cytotoxity.It suitable to clinical application.2.The results indicated that the conjugated Xeno-ADM could induce marked immunoinflammatory reaction involving in cytoinununity(lymphocytes and monocytes a macrophage-based) and in humoral immunity at early stage of transplantation.3.So we think that the basement membrane is related to the antigen of xeno-ADM.The removed basement membrane xeno-ADM has more advantage over the retained integrity basement membrane when transplanted as a kind of soft filling material.4.Xeno-ADM present good biocompatibility,not obvious degradation,no infections, no granuloma formation,sclerosis implanted in rats.Its stable performance after transplantation promote Xeno-ADM to play a role of new soft tissue filing material.
