PARTⅠEFFECTS OF SLEEP DEPRIVATION AND RIVAL ON SPATIAL MEMORY OF RATS AND MATRIX METALLOPROTEINASE-9 AND MODAFINIL INTERVENTIONObjective:To investigate the change of spatial memory of rats and matrix metalloproteinase-9 and tissue matrix metalloproteinase-1 after various degrees of sleep deprivation and sleep revival and modafinil intervention.To discuss the effect of matrix metalloproteinase-9 on spatial memory impairment and synaptic plasticity after sleep deprivation and modafinil intervention.Methods:All the male SD rats were divided into 3 groups randomly:cage control group(CC),tank control group(TC) and rapid eye movement(REM) sleep deprivation and revival group.REM sleep deprivation group was divided into control sleep deprivation (SD),CMC auxiliary solvent group(CMC) and modafinil administratsion group(MD). Animals in REM sleep deprivation and revival group were sacrifced at the end of 1d,2d,3d sleep deprivation periods respectively and at the end of 2h,6h sleep revival after 3d sleep deprivation.The modified multiple platform method(MMPM) was used to establish sleep deprivation model.The special memory was tested by Y-type maze.Western-blot and real-time PCR technique were specially used to analysis the quantitative level of matrix metalloproteinase-9(MMP-9) and tissue metalloprotease-1(TIMP-1) in rats' hippocampus.Results:1)There were no statistically significant differences(p>0.05) in error reaction numbers(EN) and total reaction time(TRT) of TC,MD and R6h groups.Compared with CC,EN of SD was increased in S2d and S3d(p<0.05),TRT of SD was no significant difference.2) Compared with CC and TC groups,the quantitative of MMP-9 and TIMP-1 were significantly increased(p<0.05).Compared with CMC group at the same time,the quantitative of MMP-9 and TIMP-1 were significantly decreased(p<0.05) in MD and sleep revival groups.Conclusion:1) Spacial memory could have been adverse effect and the quantitative of MMP-9 and TIMP-1 were significantly increased in SD group,decreased in MD and R6h groups after REM.sleep deprivation.2) Modafinil and sleep revival may improve the memory after sleep deprivation. Animals in REM sleep deprivation and revival group were sacrificed at the end of 1d,2d, 3d sleep deprivation periods respectively and at the end of 2h,6h sleep revival after 3d sleep deprivation.The modified multiple platform method(MMPM) was used to establish sleep deprivation model.The special memory was tested by Y-type maze.High pressure liquid chromatography technique were used to analysis the quantitative of DL-Aspartic acid(Asp),L-Glutamic acid(Glu),Glycine(Gly) andγ-Aminobutyric acid(GABA) in rats' hippocampus.Results:1)There were no statistically significant differences(p>0.05)in error reaction numbers(EN) and total reaction time(TRT) of TC,MD and R6h groups.Compared with CC,EN of SD was increased in S2d and S3d(p<0.05),TRT of SD was no significant difference.2) Compared with CC and TC groups,the quantitative ofβ-dystroglycan were significantly increased(p<0.05).Compared with SD group,the quantitative ofβ-dystroglycan were significantly decreased(p<0.05) in MD and R6h groups.3) Compared with CC and TC,the quantitative level of Asp and Glu were significantly increased(p<0.05),the quantitative of Gly were significantly decreased.Compared with CMC group at the same time,the quantitative level of Asp and Glu were significantly decreased(p<0.05),Gly were significantly increased in MD and sleep revival groups (p<0.05).The quantitative of GABA were not significantly regular in SD and MD groups。Conclusion:1) Spacial memory could have been adverse effect and the quantitative level of inhibitory amino acidsneurotransmitter were significantly increased and excitatory amino acidsneurotransmitter were significantly decreased in SD group.Inhibitory amino acidsneurotransmitter were significantly decreased and excitatory amino acidsneurotransmitter were significantly increased in MD and R6h groups after REM.sleep deprivation.2) Modafinil and sleep revival may improve spatial memory of rats after sleep deprivation through effect on releasing of amino acidsneurotransmitter.
