OBJECTIVES:The relationship between fat and adrenal hormones is very close.Fat could affect adrenal tissue with adipic factors.This study aims to compare the mRNA expression of the Long Leptin Receptor(Ob-Rb),Adiponectin receptor 1 and 2(AdipoR1/2) in human adrenal tissues and tumors.METHODS:1.Total RNA was exracted with TRIZOL from 6 normal human adrenal cortex,6 normal human adrenal medulla,14 adrenal aldosteronomas,10 cortisol-secreting tumors,20 pheochromacytomas;2.After reverse transcription,Ob-Rb,AdipoR1/2 and their internal reference GAPDH were respectively amplified;3.The mRNA levels of the target genes were semi-quantified by dividing the grayness value of target band by that of its reference band,represented by Ob-Rb/GAPDH,AdipoR1/GAPDH, AdipoR2/GAPDH,respectively.The ratios of each target genes were compared between different adrenal tissues.RESULTS:1.The mRNA of Ob-Rb and AdipoR1/2 were expressed in normal human adrenal cortex,normal human adrenal medulla,adrenal aldosteronomas,cortisol-secreting tumors and pheochromacytomas;2.The mRNA expressions of Ob-Rb/GAPDH were significantly higher in normal human adrenal cortex than in all the other tissues(p<0.05),while the mRNA expressions of Ob-Rb/GAPDH in aldosteronoma were higher than cortisol-secreting tumors.The ratios of Ob-Rb/GAPDH were as followed:normal human adrenal cortex(0.32±0.12),normal human adrenal medulla(0.07±0.03 p<0.01 Vs C),adrenal aldosteronomas(0.15±0.10 p<0.01 Vs C),cortisol-secreting tumors(0.05±0.02 p<0.05 Vs APA,p<0.01 Vs C) and pheochromacytomas(0.14±0.09 were),respectively. 3.The mRNA expressions of AdipoR1/GAPDH were significantly lower in normal human adrenal cortex than in aldosteronoma and cortisol-secreting tumors(p<0.05).The mRNA expressions of AdipoR1/GAPDH in pheochromacytomas were higher than normal human adrenal medulla.The ratios of AdipoR1/GAPDH were as followed:normal human adrenal cortex(0.30±0.03),normal human adrenal medulla(0.17±0.04 p<0.01 Vs C),adrenal aldosteronomas(0.58±0.22 p<0.01 Vs C), cortisol-secreting tumors(0.48±0.13 p<0.01 Vs C) and pheochromacytomas(0.49±0.25 p<0.05 Vs M),respectively.4.The mRNA expressions of AdipoR2/GAPDH were significantly lower in normal human adrenal cortex than in aldosteronoma and cortisol-secreting tumors(p<0.05).The mRNA expressions of AdipoR2/GAPDH in aldosteronoma were lower than cortisol-secreting tumors.The ratios of AdipoR2/GAPDH were as followed:normal human adrenal cortex(0.41±0.13),normal human adrenal medulla(0.20±0.09p<0.05 Vs C),adrenal aldosteronomas(0.60±0.17 p<0.05 Vs C), cortisol-secreting tumors(0.84±0.34 p<0.05 Vs APA,p<0.05 Vs C) and pheochromacytomas (0.31±0.12),respectively.5.The total of pheochromacytomas were devided into 2 groups with the methods:benign and malignant;pheochromocytoma and paraganglioma;familial and sporadic.And there were not significantly different in these groups.CONCLUSIONS:1.The mRNA of Ob-Rb and AdipoR1/2 were expressed in normal human adrenal cortex,normal human adrenal medulla,adrenal aldosteronomas,cortisol-secreting tumors and pheochromacytomas.2.The mRNA expressions of Ob-Rb were significantly higher in normal human adrenal cortex than in all the other tissues(p<0.05),while the mRNA expressions of AdipoR1 and AdipoR2 were higher in aldosteronoma and cortisol-secreting tumors than in normal human adrenal cortex (p<0.05). 3.The mRNA expressions of AdipoR1/GAPDH were significantly higher in pheochromacytoma than in normal adrenal medulla(p<0.05).It means that Lepin and adiponectin may play a role in adrenal tumors. CONCLUSIONS:1.The protein of Ob-Rb were expressed in normal human adrenal cortex,normal human adrenal medulla,adrenal aldosteronomas,cortisol-secreting tumors and pheochromacytomas.2.The protein expressions of Ob-Rb/β-ACTIN were significantly higher in normal human adrenal cortex than in all the other tissues,while the protein expressions of Ob-Rb/β-ACTIN in pheochromacytomas were higher than normal human adrenal medulla.This result confirmed the result of mRNA in the former part,and it means that leptin may play a role in adrenal tumors. OBJECTIVES:This study aims to value the plasma level of leptin in primary aldosteronism and pheochomocytoma.METHODS:Plasma was kept from 20 controls,15 pheochromocytomas,15 aldosteronomas,14 idiopathic aldosteronism and 11 cortisol-secreting tumors,The plasma level of leptin was measured by ELISA and compared each other.RESULTS:1.The plasma levels of leptin were not significantly different between primary aldsteronism and nomal control.The plasma levels of leptin were also not significantly different between aldosteronomas and idiopathic aldosteronism.2.The plasma levels of leptin were not significantly different between pheochromocytoma and nomal control.The plasma level of leptin in pheochromocytoma and normal control was 1.61(0.65, 3.22)ng/ml and 1.16(0.74,4.85) ng/ml,respectively.3.The plasma levels of leptin were significantly higher in cortisol-secreting tumors than in normal control.The plasma level of leptin in cortisol-secreting tumor and normal control was 50.00(31.74, 50.00) ng/ml and 13.89(7.57,16.69) ng/ml(p<0.01),respectively.CONCLUSIONS:The plasma levels of leptin were not significantly different between primary aldstronism and nomal control.The plasma levels of leptin were not significantly different between pheochromocytoma and nomal control.The plasma levels of leptin were significantly higher in cortisol-secreting tumors than in normal control.It means that glucocorticoid may play a role in the secretion of the leptin.
