OBJECTIVES:VEGF is closely relevant to the growth and metastasis of tumors. The proliferation of cells and angiogenesis of tumors are regulated by AngII directly or other cytokines . But there were few reports about the effects of VEGF and AngII on the growth of adrenal tumors. Therefore, we investigate VEGF, AT1 and AT2 expression in the adrenal tumor tissues and measure the MVD of tumor stroma by counting the number of microvessle through CD34 staining by immunohistochemical method, combining with clinical materials , to explore the significance of above factors in the growth and development of adrenal tumors. METHODS:1. The specimens of 59 adrenal tumors operated from 1990 to 2005 were obtained from PUMC, including 40 cases of PHEO (benign 27, malignant 13 cases ), 10 cases of APA and 9 cases of CS, 8 cases of normal adrenal cortex and 5 cases of adrenal medulla beside tumor were controls.2. Immunohistochemistry methods: Envision two steps is used for CD34 staining, SABC method used for VEGF, AT1, AT2 staining.3. Semi-quantitative point scores for VEGF,AT1,AT2 staining: Calculated as the percentage and intensity of staining : low expression wasdefined as 0~3 point scores, and high expression was 4~6 point scores. MVD is calculated the average counts of positive CD34 staining under three unrepeated 200X high power field4.
