| Belamcanda chinensis (she-gan) is one of the commonly used Traditional Chinese Medicines (TCM). In the clinical practice, the famous medicine has been used to treat gula affection for a very long period of time and has produced quite a good effect.Exact pharmacologic action was studied in this paper. Related effects of Belamcanda chinensis were selected as pharmacological index. Investigating the inhibition effect of Belamcanda chinensis extracts on voix pedis swelling of rats; study of the Belamcanda chinensis extracts on external ear swelling of mice induced by dimethyl benzene; the stretching frequency induced by acetic acid (analgesia experiment ); the effect on cough frequency,incubation period induced by strong aqua (stopping cough); the influence of phenol red secretory volume in trachea; the ability of carbo englobement clearance to mice RES; the formation of mice hemolysin antibody induced by chicken red blood cell; febrile induced by yeast powder and bacteriostasis effectiveness in vivo and in vitro et al. The conclusion indicated that Belamcanda chinensis has the conspicuous pharmacologic action of anti-inflammatory, stopping cough, eliminating phlegm,enhancing immune function, relieving fever, analgesia, bacteriostasis et al.Using water as the extracting solvent, the effects of bacteriostasis in vivo and in vitro, anti-inflammation, stopping cough with different polarity solvent extracts were studied; RP-HPLC method with diode-array detector was developed to obtain fringerprints of extracts with water and different solvents. Areas of 48 peaks were selected as chemical data. Stepwise regression analysis was applied to interrelate the chemical and pharmacologic data. The therapeutic material basis was defined through the effects of bacteriostasis, anti-inflammation, stopping cough in Belamcanda chinensis.Pharmacodynamics methods for therapeutic material basis of stopping cough in Belamcanda chinensis with different extraction and combination methods were established. The kinesics process of cough frequency induced by ammoniae aqua and incubation period indicated that the cough incubation period prolonged obviously after administration of extracts with hot water and had positive correlation with dosage; the cough frequency decreased obviously and had negative correlation with dosage. Mean plasma concentration-time curves were best fitted to one-compartment models. The lowest limit of available quantitation, T1/2 with effect , T1/2 with effect elimination, effective range, Tmax for incubation period were 24.83mg/kg, 0.272h, 2.579h, 4.14h, 1.0h, respectively; for stopping coughing were 5.06mg/kg, 1.314h, 8.226h, 5.73h, 1.0h, respectively. There was the same phenomenon for the alcohol extracts and the relative parameters for incubation period were 1.02mg/kg, 8.427h, 8.428h, 6.64h, 1.0h, respectively; for stopping coughing were 0.34mg/kg, 2.149h, 3.971h, 7.74h, 2.0h, respectively.After drug combinated with the therapeutic material basis of anti-inflammation and bacteriostasis, the phenomenon was the same as above. The relative parameters for incubation period were 2.12mg/kg, 0.553h, 5.259h, 7.29h, 2.0h, respectively; the relative parameters for stopping cough were 0.41mg/kg, 0.538h, 8.026h, 7.38h, 2.0h, respectively.A simple and sensitive RP-HPLC method was developed for the determination of tectoridin and tectorigenin in plasma and applied for its pharmacokinetic study in rats after administration of Belamcanda chinensis extracts. After the plasma was deproteinized by acetoacetate, the plasma samples were analyzed with external reference method. The linear ranges for tectoridin and tectorigenin in rat plasma were 0.27~8.64μg/mL (r=0.997), 1.62~51.74μg/mL (r=0.9997), respectively. The LOQs were 0.27μg/ml, 1.62μg/ml, respectively. The average absolute recoveries of tectoridin at low, medium and high concentrations were 89.2%,85.8%,94.6 % .And tectorigenin were 82.9%,74.9%,81.0 %.After Belamcanda chinensis extracts were orally administrated to rats, the mean plasma concentration-time curves of tectoridin and tectorigenin were plotted and the pharmacokinetic parameters were calculated. The pharmacokinetic parameters for tectoridin and tectorigenin after administration of extracts were in the following that Tmax, Cmax, T1/2 and AUC0-∞for tectoridin were 1.5h, 0.89μg·mL-1, 1.05h and19.769 mg·h·mL-1, respectively; for tectorigenin were 1.5 h, 3.35μg·mL-1, 8.863h and 23.927mg·h·mL-1.At the guide of the theory and clinical practice of TCM, the therapeutic material basis was elucidated in this paper. Pharmacokinetic study of tectoridin and tectorigenin were carried out in rats to take a limited view of pharmacokinetic profiles of Belamcanda chinensis. This research provided a significant exploration for the modernization of Chinese traditional medicine. |
