The modern analytical technique is applied to the research offingerprint by using figures to describe the internal chemicalinformation of the traditional Chinese medicine. The fingerprint canreflect the different of description and quantity of chemicalinformation. It can be used to control the quality of the traditionalChinese medicine and drugs preparation through the full-scale andintegrative Chinese medicine which could be ratified in theinternational.Belamcanda chinensis (L.)DC. is a medical plant from Belamcanda genus, Iridaceae. There are only one species in our country. Belamcanda chinensis(Iridaceae) is a perennial shrub, distributed on the warm hillsides, edgeof the forest and meadow where have enough sunshine in East Asia includingKorean, peninsula. In China, the plants distributed in the whole countryeverywhere, whose rhizomes have been used as Chinese traditional medicineto treat flu, cough, inflammation, asthma, pneumonia as well as throattroubles such as tonsillitis, pharyngolaryngitis, laryngeal edema and soon. As one of the Chinese herbal medicines commonly used, Belamcandachinensis can counteract poison to swallow, dispel phlegm and promoteblood flow to decrease throat swelling.Studies on the chemical constituent of B. chinensis investigated thatthere are flavonoids, ketone, triterpenoid, steroid and some othercomponents. Modern pharmacological research shows that the isoflavonescontain in it have anti-inflammatory, anti-virus, anti-oxidation, anti-tumor effect and female-like effect, can selectively treat andprevent of cardiovascular diseases. Therefore it is meaningful to develop and utilize it. The tectoridin,iridin,dichotomitin,irigenin andirisflorentin which were used as the identification reference to theFingerprint in this paper belong to isoflavones. The mangiferin belongsto xanthone.A large polar compounds was extracted from the acetoacetate site ofthe rhizoma Belamcandae, which performance to yellow amorphous powder. Onthe base of chemical and spectral (UV, 1HNMR, 13HNMR) and by Checking withthe literature, the compounds was identified as 2-C-β-D-glucopyranosyl-1, 3, 6, 7-tetrahydroxy xanthone (Mangiferin, C19H18O11).The preparation method of the sample solution for the fingerprint ofthe B. chinensis has been studied, and two mobile phase systems—MEOH-H2Oand acetonitrile-H2O were considered. For the purpose of optimize themobile phase systems, we added phosphoric acid, dimethyl carbinol andtetrahydrofuran to improve the peak shape and resolution, we studied theconcentration of them at the same time, then decided that the mobile phasesystem was A: acetonitrile—B: 0.1%dimethyl carbinol and 0.1%phosphoricacid. The mobile phase gradient of elution is 0~15min: acetonitrile10%~18%; 15~40min: acetonitrile 18%~20%; 40~100min: acetonitrile 20%~100%; 100~110min: acetonitrile 100%~10%. The temperature of the columnwas 40℃, and The volume of sample was 5μl. According to the DAD detectorwe chose 265nm as the detection wavelength, Identified six peaks in thefingerprints of the rhizoma Belamcandae they were Mangiferin,tectoridin,iridin,irigenin,irisflorentin and dichotomitin.After research technique of B. chinensis fingerprint were reviewed inAdaptability Test of System and method for the validation, HPLCfingerprint of rhizoma Belamcandae was established, The method is stable, reliable and had good reproducibility. Fingerprints showed theMangiferin,tectoridin,iridin and irigenin are all single element, therefore their content can be Determinationed through external referencemethod. The HPLC fingerprint can be used as the quality control methodsof the Compounds as tectoridin in the rhizoma Belamcandae, it would bemore scientific and comprehensive than only determinate the content ofthe irisflorentin which was Included in the 2005 version ChinesePharmacopoeia. We collected 16 origin rhizoma Belamcandae, and selectedthe sample from Tuanfeng GAP base as a standard to establish theFingerprints standards, chose the peak of tectoridin as the reference peakto Calculate the relative retention time and relative peak area of thecommon peaks. Calculated the similarity of the fingerprints of rhizomaBelamcandae from different origin, Using three different methods ascorrelation coefficients,cosine angle and Euclid distance, thusgenerally arriveat the different qualities of rhizoma Belamcandae fromthe different origin. These researches may lay the foundation for the studyof germplasm resources of Belamcanda chinensis (L.)DC., and Provide animportant scientific basis to the development and utilization of theresources of it.
