Studies On Banxia Baizhu Tianma Decoction By Semi-Bionic Enzyme Extraction Mode

Objective:Taking Banxia Baizhu Tianma Decoction as an example to probe into the basic mode on extraction of available compound substances in Chinese herbal medicine by semi-bionic enzyme extraction(SBEE).Methods:First,using orthogonal design to optimize SBEE technical conditions for Banxia Baizhu Tianma Decoction,with gastrodine,glycyrrhe-tinic acid,total area of HPLC,extracts(≤1000Da)polysaccharide and dry extract as comprehensive evaluated indexes,In addition,optimizing the combined ingredients of extraction methods of Banxia Baizhu Tianma Decoction.Next,using homogeneous design to optimize SBE technical conditions for Banxia Baizhu Tianma Decoction,and using proportional method to optimize the alcohol concentration for extraction.Next,comparing the HPLC fingerprint,rabbit serum containing fingerprint,effects on rats of dampness type hypertension,effects on experimental animals of resistance type dampness in the inner ear vertigo(Meniere's syndrome),maximum tolerated dose of the 4 kinds of solution(SBEE,SBE,WE,AE),and comprehensive analysis the results to judge whether SBEE was the best Extraction method for Banxia Baizhu Tianma Decoction.Results:①The optimized SBEE extraction conditions were the following:First pretreating herb with 2%enzyme I of the total weight for 12h,pH values of the water for the thrice estraction being 2.0,6.5,9.0,adding 0.5%enzymeⅢto the first extraction solvent,adding 1.0%enzymeⅣto the third extraction solvent,and thrice extraction time being 60min,30min,30min and the temperature of extraction being 61℃±1℃.②The optimal combination of this prescription being for herb Rhizoma Gastrodiae and herb Rhizoma Pinelliae,herb Poria and herb Exocarpium Cirri Rubrum,herb Largehead Atractylodes Rhozome and herb Licorice Root to be decocted respectively before the decoction was combined finally.③The optimized SBE extraction conditions were the following:pH values of the water for thrice extraction being 4.0,6.5,8.0,and extraction time being 120min,90min,90min.④the optimized alcohol concentration for extracting Banxia Baizhu Tianma Decoction being 30%.the order of comprehensive evaluated values of the 4 kinds of extracting solution being Y_SBEE＞Y_SBE＞Y_WE＞Y_AE.⑤Four kinds of extracting solution of the total area of characteristic peaks in HPLC fingerprint being not equal,the total area of SBEE liquid being the largest.⑥Comparisoning four Ways to extract of rabbit blood serum containing fingerprint on the same moment,Y of SBEE solution was the largest.⑦Comparisoning effects of four kinds of extracting solution on dampness hypertension rats,on experimental animals in the Meniere's syndrome,SBEE was best.In comprehensive analysis SBEE was better.In the experiment of maximum tolerated dose in mice,experimental animals were not death,showing that extracts of various ways were safe and reliable.Conclusions:comparing multicomponent with SBEE solution with SBE solution,WE solution and AE solution in this study,all the results showed that SBEE solution was the best.The established semi-bionic enzyme extraction patterns was a new way of thinking and method for compound substances in solution of Chinese Medicines.
