Studies On Banxia Baizhu Tianma Decoction By Semi-Bionic Enzyme Extraction Mode (Ⅰ)

Objective:Take Banxia Baizhu Tianma Decoction as an example to probe into the basic mode on extraction of available compound substances in Chinese herbal medicine by semi-bionic enzyme extraction(SBEE).Methods:First,using orthogonal design to optimize SBEE technical conditions for Banxia Baizhu Tianma Decoction,with gastrodine,glycyrrhe -tinic acid,total area of HPLC,extracts(≤1000)polysaccharide and dry extract as comprehensive evaluated indexes,ln addition,to optimize the combined ingredients of extraction methods of Banxia Baizhu Tianma Decoc -tion.Next,using homogeneous design to optimize SBE technical conditions for Banxia Baizhu Tianma Decoction,and using proportional method to optimize the alcohol concentration for extraction.Next,compare the 4 kinds of solution of SBEE method,SBE method,WE method,AE method on compositions and comprehensive analysis the results to judge whether SBEE is the best extrac -tion method for Banxia Baizhu Tianma Decoction.Results:The optimized SBEE extraction conditions are the following:First pretreating herb with 2%enzyme I of the total weight for 12h,PH values of the water for the thrice estraction is 2.0,6.5,9.0,added 0.5%enzymeⅢto the first extraction solvent,added 1.0%enzymeⅣto the third extraction solvent,and thrice extraction time is 60min,30min,30min and the temperature of extraction is 61±1℃.The optimal combination of this prescription is for herb Rhizoma Gastrodiae and herb Rhizoma Pinelliae,herb Poria and herb Exocarpium Cirri Rubrum,herb Largehead Atractylodes Rhozome and herb Licorice Root to be decocted respectively before the decoction iS combined finally. The optimized SBE extraction conditions are the following:pH values of the water for thrice extraction is 4.0,6.5,8.0,and extraction time is 120min,90min,90min;the optimized alcohol concentration for extracting Banxia Baizhu Tianma Decoction is 30%;the order of comprehensive evaluated values of the 4 kinds of extracting solution is Y_SBEE>Y_SBE>Y_WE>_Y(AE).Conclusion:this study carried out the compare of multicomponent for SBEE solution with SBE solution,WE solution and AE solution,all the results show that SBEE solution is the best.The established semi-bionic enzyme extraction patterns provide a new way of thinking and method for compound substances and development for New Chinese Medicines.
