Effect Of DAT Upon The Expression Of BFGF In Retinal Pigment Epithelium Of Guinea Pigs With Form-deprivation Myopia

Objective To explore the expression of dopamine transporter(DAT) and basic fibroblast factor(bFGF) in retinal pigment epithelium of form deprived guinea pigs.Method Form deprivation myopia in guinea pigs was induced by occlusion of the right eyes with translucent goggles. Thirty guinea pigs were divided into 3 groups:group A-normal control group, group B-the right eyes of the guinea pigs weared a translucent goggles for two week, and group C-the right eyes of the guinea pigs weared a translucent goggles for the first week. After 14 days of goggles occluding, myopia was confirmed by optometry, axial length was measured by A ultrasonography and corneal curvature was measured by photokeratometer. Subsequently, dopamine transporter protein of retinal pigment epithelium was evaluated by the immunohistochemistry and Western blotting, and the mRNA of dopamine transporter was evaluated by RT-PCR. Basic fibroblast factor protein in retinal pitment epithelium was evaluated by Western blotting. The content of dopamine in retinal pigment epithelium was evaluated by high performance liquid chromatography-electrochemical detection (HPLC-ECD).Results The diopter is+0.917±0.303 D in group A,-3.541±0.579 D in group B, and+0.750±0.223 D in group C on average. The average axial length is 6.663±0.176 mm in group A,8.045±0.318 mm in group B, and 6.767±0.118 mm in group C. There are significantly statistical differences between group B and group A (P< 0.05). Dopamine transporter is mainly positively expressed in retinal pigment epithelium, and is negative in choroid. The expression of dopamine transporter is strongly positive in group B. There is a downregulation of dopamine transporter in retinal pigment epithelium in group B (P< 0.05), and an upregulation of basic fibroblast factor in retinal epithelium in group B (P< 0.05). There are positive correlations between Western blotting results and immunohistochemistry results of dopamine transporter (r=0.893, P< 0.05). There are positive correlations between Western blotting results of dopamine transporter and HPLC results of dopamine (r=0.902, P<0.05). There are negative correlations of Western blotting results between dopamine transporter and basic fibroblast factor (r=0.893,P<0.05).Conclusion 1. Dopamine transporter and basic fibroblast factor proteins are positively expressed in retinal pigment epithelium. Dopamine transporter is negative in choroid.2. There is a downregulation of dopamine transporter and an upregulation of basic fibroblast factor in retinal pigment epithelium of guinea pigs.Objective To explore the effect of dopamine transporter on the expression of basic fibroblast factor in retinal pigment epithelium in form-deprived guinea pigs through using exogenous dopamine transporter antisense oligonucleotides.Method Form deprivation myopia in guinea pig was induced by occlusion of the right eyes with translucent goggles. Fifty guinea pigs were divided into 5 groups:group A-normal control group, group B-the right eyes of the guinea pigs weared a translucent goggles for two week, group C-. the right eyes of the guinea pigs were vitreally injected with dopamine transporter antisense oligonucleotides (0.02nmol/ul), group D-the right eyes of the guinea pigs were vitreally injected with dopamine transporter sense oligonucleotides (0.02nmol/ul), and group E-the right eyes of the guinea pigs were vitreally injected with saline solution. After 14 days of goggles occluding, myopia was confirmed by optometry, axial length was measured by A ultrasonography and corneal curvature was measured by photokeratometer. Subsequently, dopamine transporter protein of retinal pigment epithelium was evaluated by Western blotting and the mRNA of dopamine transporter was evaluated by RT-PCR. There are significantly statistical differences between group B and group C (P< 0.05). Dopamine transporter antisense oligonucleotides has a downregulation effect upon dopamine transporter and an upregulation effect on basic fibroblast factor in retinal pigment epithelium of guinea pigs. There are negative correlations of Western blotting results between dopamine transporter and basic fibroblast factor (r=-0.973, P<0.05)Conclusion 1. Exogenous dopamine transporter antisense oligonucleotides could inhibit form deprivation myopia of guinea pigs, and has a downregulation effect upon dopamine transporter and an upregulation effect on basic fibroblast factor in retinal pigment epithelium of guinea pigs.2. The regulation effect of dopamine transporter upon form deprivation myopia might causes by the regulation of basic fibroblast factor in retinal pigment epithelium of guinea pigs.
