OBJECTIVES:To study the RIZ1 mRNA,which gene was selected form the our formal meningioma cDNA microarrays analysis, expression and distrbution in the meningioma tissue. To resech the role and possible mechanism of RIZ1 gene in malignancy progress of meningiomas. METHODS:Applications Real time-PCR and tissue microarray immunohistochemical research RIZ1 at all levels of WHO in the distribution of meningioma specimens. To establish RIZ1 gene over-expression cell model by transfecting the completed plasmid RIZ1 to the IOMM-LEE cell line, a malignant meningioma cell line. RIZ1 gene expression observed after IOMM-LEE cell proliferation and to discuss relation with the proto-oncogene c-myc expression.The use of methylation-specific PCR to detect methylated RIZ1 mRNA expression in benign and malignant meningiomas. Take a initial discussion for the role of RIZ1 gene involved in mechanism of meningioma malignant proliferation.RESULTS:The RIZ1 mRNA and protein has differential expression between the meningioma pathology grades of WHOâ… ,â…¡andâ…¢and with decreased its expression compare to the tumor grade increased. RIZ1 gene overexpression in malignant meningioma cells IOMM-LEE significantly inhibited the proliferation and with c-myc gene expression was negatively correlated. In the WHOâ… ,â…¡andâ…¢level of meningioma specimens were detected by DNA Methylation of the RIZ1 gene, and increased with the grade, were found the higher the proportion of methylation RIZ1mRNA.CONCLUSIONS:RIZ1 gene expression exist in meningiomas and, their distribution was negatively correlated with the degree of malignant meningioma. RIZ1 gene with a clear function of inhibiting the proliferation of malignant meningiomas. RIZ1 associated with the control mechanism is subject to certain factors have made it possible because of RIZ1 was methylated RIZ1 in turn lost its inhibitory effect of c-myc by c-myc proto-oncogene expression increases contributed to the proliferation of meningioma cells...
