Marine microorganisms have become an important source of pharmacologically active metabolite. Marine-derived fungus was one of the important members, and it was value to exploit potence. Mangrove marine-derived fungus could produce numerous novel and bioactive compounds, so it has an expansive foreground to study. This dissertation included two sections in five chapters.The first part (chapter 1) is a review on the advance of bioactive metabolites of marine fungi. Novelty metabolites are introduced from Marine microorganisms and phytoplankton in the near years. This review mainly picked up about one hundred and forty-two new compounds from literaturel and their bioactive research .The second part(chapter 2 to chapter 5, body part)is our studies on the metabolites of six mangrove fungi (No. No. ZSU-h76,ZH-111,SK7RN3G1,5094,GX2-1C,ZH-58) collected from the South China Sea. 50 compounds, including 4 new compounds, 2-1,2-2,2-3 and 3-1 were isolated from these fungi through chromatographic separation. Their structures were identified by UV, IR, NMR, MS and so on. Compound 2-1, 2-2 and 2-3 are all dimmers, 2-2 and 2-3 possesses a pyrano[4,3-b]pyran-5(2H)-one ring system that is unprecedented in nature. Prelimin- ary pharmacological test revealed that the new compounds exhibited little cytotoxic activity towards KB, KBv200 and MCF-7 cell lines. In the antibacterial test, they showed no effect against Staphylococcus aureus, Bacillus subtilis, Escherichia coli and Sarcina lutea at 50μg/mL. But 2-1and 3-1 can accelerated the growth of subint- estinal vessel plexus (SIV) branch markedly by zebrafish embryos collection test.Chapter 2 is study on the metabolites of mangrove marine-derived fungus Phomo psis sp.zsu-H76 from the South China Sea. Three new compounds: phomopsis-H7(2-1),phomopsis-H76 B(2-2),phomopsis-H76 C(2-3),eight known compounds: cytochalasin H(2-4),mannitol(2-5),cyclo-(Phe-Gly)(2-6),benzeneacetic acid(2-7),cyclo-(Leu-Gly)(2-8), Chromone(2-9),pyridin(2-10)and ergosterol(2-11),were isolated from the fungus. Preliminary pharmacological test revealed that three new compounds exhibited little cytotoxic activity towards KB, KBv200 and MCF-7 cell lines. A new isocoumarin, (3R,4S)-3,4-dihydro-4,5,8-trihydroxy-3-methyl-1H-2-Benzopyran-1-one(3-1),togethr with 11 known compounds ,4-(Hydroxymethyl)-7-methoxy-6-methyl-1(3H)-isobenzo furanone(3-2),Exumolide A(3-3),Dilactone(3-4),4-hydroxymellein(3-5),5,8-dihydroxy- 4-methyl-coumarin(3-6),3,4-dihydro-4,8-dihydroxy-3,5-dimethyl isocoumarin(3-7),3,4- dihydro-8-hydroxy-3-methyl isocoumarin-5-carboxylic acid(3-8),3,3'-Dihydroxy-5,5'-dimethyldiphenyl ether (Diorcinol)(3-9),5-methyl- 2,4(1H,3H)-Pyrimidinedione(3-10),Fusaric acid(3-11),3,5-dihydroxy ergosta-7,22- diene-6-keteon(3-12). Primary bioassays showed that compound (3-1) and (3-3) can accelerate the growth of blood vessel markedly and compound (3-2) can inhibit the growth of blood vessel. Three compounds(3-1,3-2,3-3)exhibited weak cytotoxicity against Hep-2 and HepG2 cells (IC50>50 mg/mL).Chapter 4 is study on the metabolites of mangrove marine-derived fungus (SK7 RN3G1) from the South China Sea.12 compounds, 1,3,8-trihydroxy-6-methyl-9,10- anthraquinone(4-1),Lumichrome (4-2),Curvularin (4-3),physcion(4-4),emodindi-meth ylether(4-5),Chrysophanol (4-6),1,4,6-trihydroxy-2-methoxy-7-methylanthracene -9,10-dione (4-7),emodin(4-8),3,6-di-sec-butyl-1,4-dihydroxypiperazine-2,5-dione (4-9),three knowncyclodipeptides (4-10, 4-12),cyclo-(Pro-Gly) (4-11),were isolate d from the mangrove endophytic fungus.Chapter 5 is study on the metabolites of mangrove marine-derived fungus (5094, GX2-1C, ZH-58)from the South China Sea. 15 compounds, 5,5'-oxy-dimethylene- bis(2-fur-aldehyde)(5-1), deClrorogriseofulvin(5-2), 3,8-dihydroxy-6-methyl-9-oxo- 9H-xanthene-1-carboxylate (5-3), lichenxanthone (5-4), Griseoxanthone C (5-5), 1,3, 6-trihydroxy-8-methyl-9H-xanthen-9-one(5-6),Harman (5-7), N-methyl- harman (5-8), three known sterol derivates(from 5-9 to 5-11), 4-hydroxy-benzoic acid (5-12), 4-hy- droxylbenzeneacetic acid(5-13), succinic acid(5-14), piliformic(5-15), were isolated from the fungus.
