To Study The Effects Of Arsenic Trioxide On The Proliferation And Apoptosis Of Human Triple Nagetive Breast Carcinoma HCC1937 Cell Line In Vitro

Purposes:To study the effects of arsenic trioxide on the proliferation and apoptosis of human triple nagetive breast carcinoma HCC1937 cell line in vitro.Methods:1.The inhibitory effects were evaluated by MTT assay.2.Fluorescence microscope was used to observe the morphology of cell apoptosis.3.The rates of cell apoptosis were detected by flow cytometry.4.The changes of Ki-67, Bax and Bcl-2 in mRNA and protein levels were quantified by RT-PCR and Western blot.Results:The growth inhibition of triple nagetive breast cell was dose-dependent obvious after exposure to the arsenic trioxide which was divided into different concentrations groups. With the arsenic trioxide concentrations rising and the period lengthening, the more increasingly the arsenic trioxide effected. All the treatment groups were observed much more early apoptosis cells than the control group under the fluorescence microscope. The inhibitory rates of the arsenic trioxide at IC50, The control group was observed less early apoptosis cells than the 24h,48h and 72h group under the fluorescence microscope. The cells apoptotic rates tended to grow in the increase order of the control,24h,48h and 72h group. Compared with the control group, any of the treatment group's Ki-67 and bcl-2 mRNA (protein) expression were greatly decreased but the expression of Bax mRNA (protein) was largely increased.Conclusion:The poisonous chinese medicine,arsenic trioxide was proved to be effective in the inhibition of the proliferation and the induction of the apoptosis of human triple nagetive breast carcinoma HCC1937 cell line and with the arsenic trioxide concentrations rising and the period lengthening, the more obviously the arsenic trioxide treated.
