To develop high-tech industry has become one of the most important national strategic policies for many countries under the current situation of global competition. The principle of "making Independent innovations and leapfrog advances in key areas, supporting development and creating a better future" has become guidelines on Building an innovation oriented country in China. Therefore, planning of the science park comes to be an important field as well as scientific frontier of urban and regional planning.Begin with studying the spatial development history of international science park, the dissertation used "question orientated approach", with the aim of urban sustainable development, Through the discussion on the planning and design strategy of the knowledge era, summarized, analyzed, and compared realistic problems of Nanjing area, studied major conflicts and related policies under the backdrop of urban transformation and restructuring in China.1) Summarized process and characteristics of physical planning, related concepts, developing rules, and scientific theories of university science parks in the world. Discussed the experiences and lessons to learn of science park planning and construction.2) Introduced concept and characteristics of "knowledge city". Discussed the relevancy between University Science Park and knowledge city from the aspects of innovation network, industry development, location and so on.3) Summarized the planning strategy of the university science park and integrated design strategy of the Research and Development Building. With case study, comprehensively analyzed the influence of park planning and architecture design to the innovation spatial development and vice versa.4) Analyzed major problems of spatial development in Nanjing area. Discovered the main contradiction we are facing during development and physical planning of Nanjing and university related science parks. Proposed that the imbalanced policies and massive real estate development have negative impacts on Nanjing’s innovation environment and social structure; the chaotic concepts of the architecture design and systematic weakness of current city planning cannot meet the challenge of city development; the physical planning of Nanjing and university related science park needs new concepts and arrangements.5) Based on analyzing fundamental problems of Nanjing science park’s physical planning, following the master planning of Nanjing region, referencing the successful experiences of science parks in the world, proposed the following: first, the development of the university related science park needs to be viewed as regional and urban planning; second, the city physical planning should consider the various possibilities of city development, not single model development; third, economy, politics, nature, society, and culture space are not completely different concepts, they are connected, the high priority job of city physical planning is to establish a comfortable culture environment, to provide the assurance of virtual space development; fourth, emphasis of urban culture protection and creation are necessary to establish the city physical appearance for a sustainable development of Nanjing high-tech industry; fifth, emphasis of the study to countermeasures and methods of the Research and Development Building’s design. The detailed reformation procedures are proposed as well.
