Breeding Of Genetic Male Sterile Lines In 4 Special Vegetables Of B. Campestris

In order to resolve the problem that cross breeding in utilizing hybrid vigor of 4 special types of Brassica crops in B.campestris (AA, n=10), the materials of multiple allele inherited genetic male sterile line bred by our group in previous time were used as the source of male sterility. The directional transfer scheme was designed to breed the genetic male sterile lines of the 4 Brassica crops. The main results were as follows:1. The genetic male sterile line of milk Chinese cabbage GMS3 was used as the test cross line to identify the genotype of the fertile target line’Gongguhuirui’cabbage Y02. The results showed that the genotype of the fertile target line Y02 on the multiple allele position of genetic male sterility was purified-dominant male restorer genotype MsfMsf. Directional transfer scheme was designed to breed the genetic male sterile gene from the male sterile line GMS3 of milk Chinese cabbage into the fertile target line Y02. The new genetic male sterile line A01 with its corresponding AB line type I and the temporary maintainer line was bred successfully, which was stable inheritance and similar to the fertile target line Y02 in botanical and economic characters and with 100% rate of sterility plants and 100% degree of sterility flowers. In the process of breeding, the SCAR marker syau_scrO4 which linked to the male sterility gene Ms developed by our group members was applied in the selection of the plants contained Ms gene. The selection results indicated that the accuracy of marker syau_scrO4 in determining the plants genotype was 100%, which showed that the marker could be applied in the marker-assisted selection of the genic multiple-allele male sterile line. Cross combinations were made by using the new bred male sterile line A01 as female and 8 selfing lines of different Chinese cabbage as male, carrying out the experiment of comparing the varieties.2 excellent cross combinations (D3 and D4) were selected.2. Sexual hybridization was applied to identify the genotype of the fertile target line Y0701 of dwarf Chinese cabbage with the male sterile plants in AB line AB01 of Chinese cabbage as the test cross line. According to the expression of the progenies, the target line Y0701 existed MsfMsf on the multiple allele position of genetic male sterility. Compound transfer plan was made to breed the genetic male sterile line of dwarf Chinese cabbage with the AB line AB01 and the temporary maintainer line B01 as the source of male sterility and Y0701 as the target line. The new genetic male sterile line GMS00 with its corresponding AB line type I and the temporary maintainer line was bred, which had 100% rate of sterility plants and 100% degree of sterility flowers. The new male sterile lines with 100% rate of sterility plants and 100% degree of sterility flowers in BC2 and BC4 were bred successfully with the above materials through directional transfer plan. The new male sterile lines were compared with the target line Y0701, the results showed that the horticultural characters of GMS00 was between the AB line AB01 and the target line Y0701, while the uniformity degree of GMS02 and GMS04 were superior to GMS00, and the horticultural characters of GMS04 was much similar to the target line Y0701, which reached to the effect of directional transfer. Cross combinations were made by using the newly-bred male sterile lines GMS00、GMS02 and GMS04 as female and 2 selfing lines of dwarf Chinese cabbage as male, carrying out the experiment of comparing the varieties. The excellent cross combination PB401 was selected.3. The genetic male sterile line of milk Chinese cabbage GMS3 was used as the test cross line to identify the genotype of the fertile target line’Leiyong’cabbage Y01. The fertility expression of progenies indicated that the genotype of the fertile target line Y01 on the multiple allele position of genetic male sterility was purified-dominant male restorer genotype MsfMsf. Directional transfer scheme was put forward for transferring the genetic male sterile gene from the male sterile line GMS3 of milk Chinese cabbage into the fertile target line Y01. The new genetic male sterile line A02 with its corresponding AB line type I and the temporary maintainer line was bred successfully, which was stable inheritance and similar to the fertile target line Y01 in botanical and economic characters and with 100% rate of sterility plants and 100% degree of sterility flowers.4. Directional transfer scheme was designed to breed the genetic male sterile gene from the male sterile line GMS3 of milk Chinese cabbage into Komatsuna Y05. The new genetic male sterile line A03 with its corresponding AB line type I and the temporary maintainer line was bred, which exhibited stable inheritance and similar to the fertile target line Y05 in botanical and economic characters and with 100% rate of sterility plants and 100% degree of sterility flowers.
