The malonic dialdehyde(MDA) and reactive oxygen species(ROS) in sperm could reduce the conventional quality of semen, and fracture the DNA chain of sperm, causing infertility or sperm death. In the study on cattle frozen semen, there was some protective work on semen withstand MDA and ROS, by adding different substances. However, there was not so much research on this object. In this paper, firstly, the mice were chosen to feed diet with different antioxidant for exploring the better sperm protector. After feeding mice with vitamin E, B12, C, resveratrol and melatonin, it showed that vitamin E, B12, C, resveratrol and melatonin could have much more protective effects on the sperm quality, the antioxidant enzymes activity and sperm DNA integrity. Then, according this result, there was a trial on Qinchuan male cattle to find the protector on sperm. 1.In the test section of mice, supplementation of vitamin E(VE) in diet could significantly improve the sperm motility, density, activity of hyaluronidase(HYD), acid phosphatase(ACP), acrosomal enzyme(ACE), catalase(CAT), glutathione peroxidase(GSH-px) and superoxide dismutase(SOD) in sperm(P<0.05), compared with the control group, as well as distinctly reduce the content of reactive oxygen species(ROS), malonic dialdehyde(MDA) and sperm malformation rate(SMR)(P<0.05). And the VE supplementation could have the notable protective effect on sperm DNA integrity.Meanwhile, in the Qinchuan beef cattle(Chinese domestic beef cattle) trial, it showed that the supplementation VE with 60g/d-150g/d overtly improved the percentage of sperm in grade A, the activity of HYD and ACE(P<0.05). Additionally, it was obviously the supplementation VE increased sperm motility, activity of glutathione reductase(GR), ACP and sperm acrosome integrity rate(SAIR)(P<0.05), and decreased the ROS level in sperm with 20g/d-150g/d VE(P<0.05), respectively. When the VE content was between 60g/d and 150g/d, there was a distinct low level of MDA in sperm(P<0.05). Concurrently, there was a significant higher rate of sperm DNA integrity than the control with VE between 100g/d and 150g/d(P<0.05).2.In the test section of mice, it showed that supplementation vitamin B12(VB12) in diet significantly improved the SAIR, activity of CAT, SOD GSH-px and HTD(P<0.05), compared with the control. And it had reduced the ROS and MDA level in sperm, increased the sperm DNA integrity rate(P<0.05). However, there was no obvious difference at sperm quality, enzyme activity and DNA integrity(P>0.05).3.In the test section of mice, supplementation vitamin C(VC) in diet could significantly improve the activity of CAT, SOD GSH-px, HTD, ACE, ACP and sperm motility rate(P<0.05). It also increased sperm DNA integrity apparently(P<0.05). As well as, there was some notably reduced effect of MDA and ROS level in sperm(P<0.05).In the Qinchuan beef cattle trial, the diet with 6g/d VC could improve the percentage of sperm in grade B,(P<0.05), overtly, compared with the control. And in cattle group, there was a obviously higher sperm motility and lower MDA level than control(P<0.05), respective with VC in 6-10g/d and 6-15g/d. when the VC supplementation between 2g/d and 15g/d, the ROS level was below, and the GR overtopped the control significantly(P<0.05), respectively. Moreover, when the VC supplementation was 2-6g/d and 15g/d, the activity of HYD was overt higher(P<0.05). When the VC supplementation was 2g/d and 10g/d, the activity of ACE was overt higher(P<0.05). When the VC supplementation was 6g/d and 15g/d, the rate of sperm DNA integrity was overt higher(P<0.05). And when the VC supplementation was 2g/d and 10g/d, the rate of sperm DNA damage was overtly lower(P<0.05).4.In the mice trial, supplementation resveratrol in diet could significantly improve sperm motility, the activity of CAT, SOD, GR, GSH-px, HYD, AC, ACP and DNA integrity in sperm(P<0.05), respectively. Meanwhile, it showed that supplementation resveratrol could also reduce the level of MDA and ROS(P<0.05), decrease the rate of sperm deformity(P<0.05).In the Qinchuan cattle trial, analogously, supplementation resveratrol in diet could significantly improve the activity of SOD, GSH-px, GR, ACP, ACE and the rate of DNA integrity in sperm(P<0.05), respectively. Simultaneously, it decrease the MDA and ROS level of sperm(P<0.05).5. Supplementation melatonin in mice diet could significantly improve the activity of SOD, GR, GSH-px and ACP in sperm(P<0.05), respectively, notably reduce the MDA and ROS level of sperm(P<0.05).6. There also had some condition-explore work about sperm protein separation of Qinchuan cattle by two-dimensional electrophoresis. It was used to explore the optimum condition on Qinchuan cattle sperm separation, by thiourea, guanidine hydrochloride and urea- guanidine hydrochloride lysis method.To sum up, after these trials, the diet supplymented VE, VB12, VC, resveratrol or melatonin would have much more protective effects on the sperm quality and sperm DNA integrity. This was the theoretical basis on some further research on Qinchuan cattle sperm..
