Effects Of Differernt Sperm Plasma Membrane Protectant On Boar Semen Stored At Normal Temperature

In order to investigate the effect of taurine, L-Glutamine, Schisandrin B, Notoginsenoside R1 on boar semen preservation at normal temperature, four different foundation formula were used and sperm motility preservation process, effectively survival time, plasma membrane integrity and acrosome integrity were detected in this study. After screening the best foundation formula, added different concentrations of taurine(0, 0.5, 1, 5 and 10 mmol/L), L-Glutamine(0, 0.5, 1, 2, 4, 8 and 16 mmo/L), Schisandrin B(0, 1, 5, 10 and 15 mmol/L) or Notoginsenoside R1(0, 1.25, 2.5, 5, 10 and 20 mmol/L),and test semen quality, total antioxidant capacity(T-AOC) and malondialdehyde(MDA) concentration, study its effect on semen quality during preservation, determine the optimal additive amount, aims to develop new type extender of boar semen preservation at room temperature. The results of this study showed that:1.Among the four self-made extender,it includes of glucose 40 g / L,sodium citrate,6.8 g / L,sodium bicarbonate0.8 g / L, EDTA 1.5 g / L, citric acid0.3 g / L and potassium chloride 0.7 g / L. The effective survival time of extender 3(4.4 d) was significant higher than that of other extenders(P < 0.05), and after four days preservation, the sperm motility rate, plasma membrane integrity and acrosome integrity were 0.61, 55.32% and 75.12%, respectively. Followed by extender 2, the preservation effect of extender 1 and extender 4 was worser than other extenders. Therefore, the extender 3 is the best extender amount the four self-made extenders and could be used as the basis in the next stage experiment.2.After added different concentrations taurine or L-Glutamine, the result indicated: when added 5 mmol/L taurine, the sperm motility rate, acrosome integrity and the concentration of total antioxidant capacity(T-AOC) were significant higher than other groups(P < 0.05), MDA concentration was significant lower than other groups(P < 0.05).After five days preservation, the sperm motility rate, plasma membrane integrity, acrosome integrity and the concentration of total antioxidant capacity(T-AOC) and malonaldehyde(MDA) were 0.66, 47.48%, 72.10%, 2.28IU/mL and 2.63 nmol/mL in the group added 5 mmol/L taurine.The sperm motility rate,sperm plasma membrane integrity,sperm acrosome integrity in the group added 4 mmol/L L-Glutamine were significant higher than other groups(P < 0.05).After five days preservation, the sperm motility rate, sperm plasma membrane integrity,sperm acrosome integrity and the concentration of total antioxidant capacity(T-AOC) and malonaldehyde(MDA) were0.59,47.15%,74.40%,2.34 IU/mL,1.51 nmol/mL in the group added 4 mmol/L taurine.The sperm motility rate,sperm plasma membrane integrity, acrosome integrity in the group added 16 mmol/L L-Glutamine were significant lower than other groups(P < 0.05). Thus, added 5 mmol/L taurine or 4 mmol/L L-Glutamine in extender of boar sperm is the best way to increase the result of preservation in normal temperature preservation.3.After added different concentrations Schisandrin B or Notoginsenoside R1 in extender 3, the result indicated: the sperm motility rate, sperm plasma membrane integrity and T-AOC in the group added 15 μmol/L Schisandrin B in extender 3 were significant higher than other groups(P < 0.05), and MDA concentration was significant lower than other groups(P < 0.05);while there were no significant different in sperm acrosome integrity(P > 0.05) among the groups added 1,5,10,15 μmol/L Schisandrin B. After five days preservation, the sperm plasma membrane integrity and the sperm acrosome integrity were 57.10%,73.70%. The concentration of total antioxidant capacity(T-AOC) and malonaldehyde(MDA) were 2.65 IU/mL, 2.85 nmol/mL in the group added 5 mmol/L Notoginsenoside R1 after four days preservation. The sperm plasma membrane integrity and sperm acrosome integrity in the group added 5 mmol/L Notoginsenoside R1 were significant higher than other groups(P < 0.05), and the MDA concentration in groups added 1.25,2.5,5,10 mmol/L Notoginsenoside R1 were significant lower than other groups(P < 0.05). Thus, both Schisandrin B and Notoginsenoside R1 can increase the anti-oxidation ability and the quality of boar semen during normal temperature preservation, and the optimum addition concentration are 15 μmol/L and 10 mmol/L, respectively.
