Objective:To investigate the effect of tanshinone and/or electro-acupuncture on inflammation caused by ischemia. Methods:To set up the rabbit model with spinal cord ischemia reperfusion injury, twenty-four New Zealand rabbits were ischemically injured and randomly divided into four groups:1) untreated group (A,n=6);2) Tanshinone alone group (B,n=6, via intraperitoneal injection);3) electroacupuncture alone group (C,n=6, via "Yanglingquan" and "Xuanzhong"); and4) tanshinone plus electroacupuncture group (D,n=6, Tanshinone plus electroacupuncture therapy). Blood was collected at different time points:before ischemia injury;0.5;1h;4h;8h; and12h after reperfusion by rabbit femoral venous. IL-1β TNF-a IL-8IL-6IL-1Ra IL-10TNFSR I and TNFSRⅡ levels were examined as marker of inflammation. Results:Through comparing the levels of IL-1β TNF-a IL-8IL-6IL-1Ra IL-10TNFSR I and TNFSR II at time points of ischemia to those before ischemia, we observed that levels of IL-1β TNF-a IL-8IL-6IL-1Ra IL-10TNFSR I and TNFSR II increased significantly after ischemia (untreated group, P<0.05). Interestingly, we found that the treatment with Tanlla and/or electro-acupuncture inhibited the elevation of IL-1β TNF-a IL-8and IL-6(P<0.05) and enhanced the increase of level of IL-1Ra and TNFSR I (P<0.05) after reperfusion (P<0.05). Significant decrease of IL-8was observed in Tanlla plus electro-acupuncture treatment group at1h after reperfusion respectively (P<0.05).Conclusion:Level increase of IL-1β TNF-a IL-8IL-6IL-1Ra IL-10 TNFSR I and TNFSRⅡ was used as marker of rabbit inflammation model caused by ischemia. Treatments with Tanshinone alone, electroacupuncture alone, and Tanshinone plus electroacupuncture are obviously effective in improving the inflammation because the increase of level of IL-1β TNF-α IL-8and IL-6was inhibited, the increase of level of IL-IRa and TNFSR I was enhanced. Tanshinone plus electroacupuncture treatment was more effective compared with the other two treatments. Moreover, time-lapse effect was observed in Tanshinone and/or electro-acupuncture.
