Objective:To build a stable animal model of Colorectal Cancer liver metastasis; To verify genes which are possibly related to colorectal cancer and liver metastasis of colorectal cancer (LMCC) by this model.Methods:To select16suitable body weight mice(20g), about six week old. under spleen’s capsule, to inject suitable human colonrectal cancer cells, then, to support mice to8weeks, so we can see cancers on liver. So we can get a liver metastasis rate. Then, we use HCT116cells ande sw480cells to follow this model. While, we also utilize RNAi skill to make three genes silencing(SCIN, CUGBP1, CDKL1) and we utilize GFP carrier to expend slc16A7gene. Then we use the modle to verify the genes which are possibly related to colorectal cancer and liver metastasis of colorectal cancer. When we get data, we use chi square testResults:In HCT116cells, CDKL1, SCIN, SLC16A7gene have the relation of liver metastasis, In sw480cells, we find CDKL1gene and SLC16A7gene have the relation of liver metastasis, the other two genes do not get the relationship.Conclusion:1, Both in HCT116ande sw480cells, we find SLC16A7gene may have a relation of liver metastasis.2, In HCT116cells, CDKL1,SCIN gene have a relation of liver metastasis.3, In SW480cells, CDKL1gene have a relation of liver metastasis. 4,These result may are not correct, we need more evidence to verify.
