Background and Objective:Colorectal cancer (CRC) is the fourth most common malignancy in men and the third maligancy in women worldwide.Every year over one million people are diagnosed with colorectal carcinoma. Recently with the development of social economy and revolution of human diet.the incidence rates and death rates are rising during past decades. The animal model of colorectal cancer liver metastasis provides us an efficient approach for understanding the development and metastasis of colorectal cancer and evaluating new drug's effect. As a reporter gene,green fluorescent protein (GFP)can stir up green fluorescence when it is mustered by ultraviolet rays or blue lights.This research tries to compare two methods for establishing colorectal cancer liver metastasis models, which aims to provide a better method for studying the growth and metastasis pattern of colorectal cancer.Methods:(1) Establish the SW620-GFP cell line:dissociate and cultivate the tumor tissue of the SW620 cell which were transfected by GFP.(2) Injection of SW620-GFP cells under the spleen involucrum:we established the liver metastasis model through injecting 5*106 SW620-GFP cells under the spleen involucrum.(3)Surgical Orthotopic implantation (SOI) in the cecum:we established the cecum orthotopic tumor by suturing lmm3 tumor tissue under the cecal serosa.(4) Estimate the liver metastasis by using the In-Vivo Imaging System FxPro and confocal microscope.RESULTS:(1) we established the SW620-GFP cell line which emited fluorescence strongly.(2)we established the animal model of colorectal liver metastasis by injecting SW620-GFP cells under the spleen involucrum.(3)we established the animal model of colorectal cancer through the surgical orthotopic implantation (SOI) under the cecal serosa.(4)we can observe the progression of cecal tumor by In-Vivo Imaging System FxPro noninvasively,and confocal microscope can assist to estimate liver micrometastasis in vitro.CONCLUSIONS:(1) The injection of tumor cells under the spleen involucrum is a more stable and reliable method to establish animal model of colorectal cancer liver metastasis than the surgical orthotopic implantation.(2) GFP combined with confluence system to estimate the process of liver metastasis is a simple,convenient approach.
