Part1The research of the TCM syndrome rules of bone meta-stasis of breast cancer patientsObjectives:To explore the TCM etiology and pathogenesis, syndrome, the syndrome differentiation sdandard of bone metastasis of breast cancer which provide the basic research for the scientific research andclinical work.Method:Investigated110cases of clinical symptoms and tongue, pulse condition in patients with bone metastasis of breast cancer, fill out the questionnaire of bone metastasis of breast cancer TCM syndromes, using SPSS to establish the diagnostic information database, and using descriptive analysis, frequency analysis, cluster analysis for statistical analysis, based of professional knowledge, conbinating the theory of TCM, then summarize the main symptoms, and the performances of tongue and pulse.Results:1.Six types of common syndrome of bone metastasis of breast cancer were:Yangxuhanning, Qizhixueyu, Tanshiliuzhu, Ganshenyinxu, Qixuekuixu.2.The main symptoms of bone metastasis of breast cancer are local pain or soreness, continuous pain, fixed pain, emaciation, bradykinesia, lassitude, poor appetite and etal.Conclusion:Reveals the distribution regularity of TCM syndromes of bone metastasis of breast cancer which conducive to the guidance of TCM and clinical treatment. Part2The experimental studyObjectives:To investigate the effection of Yanghe decoction for the Yang deficiency syndrome of bone metastasis of breast cancer nude mice model and related its mechanism.Method:1.Intramuscular injected hydrocortisone for nude mice to established the of model Yang deficiency syndrome.2.Injected breast cancer MDA-MB-231cells suspensionto the ventriculus sinistee to established nude mice bone metastasis model.3.Using Yanghe decoction intervention treatment for bone metastasis of breast cancer nude mice model, dynamic observation of nude mice survival, the body weight changes, imaging, making the X-ray imaging, conducting the imaging score, pathology observation, determination of cAMP, cGMP, T3, and determination of OPG, RANKL level by PT-PCR and Western-blot.Results:1.After injected hydrocortisone, the nude mice had Yang deficiency performance, thus as aversion to cold, reduced activity, apathetic, fur less shiny, eating less, thin stool, and cAMP decreased, cGMP increased, cAMP/cGMP decreased, T3increased obviously.2. After injected breast cancer cells suspension to the ventriculus sinistee, the nude mice appeared different degree bone metastasis, and the X-ray imaging examination and histopathological examination confirmed the diagnosis.3.Yanghe decoction can inhibit bone metastasis to a certain extent, induced osteolytic bone destruction of bone metastasis, improve the quality of nude mice’s life, and can increase the expression of OPG, inhibited expression of RANKLConclusion:1. Hydrocortisone injection can established nude mice mode of Yang deficiency successfully.2.Injected breast cancer cells suspension to the ventriculus sinistee can established the model of bone metastasis of breast cancer in a high successful rate.3Yanghe decoction can improve the Yang deficiency symptom by adjusting the T3, cAMP, cGMP, cAMP/cGMP.4.Yanghe decoction can increase the expression of OPG, inhibit the expression of RANKL to inhibit the development of bone metastasis of breast cancer.5.Yanghe decoction’s functions may be related to promoting or inhibiting cAMP, cGMP expression.
