Objective:The present study aims to use gene chip technology searched different related gene to analyze differences of cervical cancer-changing spectrum, to validate apoptosis-related gene, explore the pathogenesis of cervical cancer of the Uighur and the relationshipwith clinicopathological parameters.Methods:Affymetrix genome-wide oligonucleotide microarray was used of expression Porfiling for5cervical cancers and5cervix without disease.The microarray contained36000transcripts/20000genes,Genes with differential expression were identified by analytic tools of bioinformatics and functional genomics.real-time quantitative PCR,immunohistochemistry were used to validate expression of genes:Bcl2、Bcl-x1、c-IAP1in45carvical cancer and21cervix without disease.Analysis the relationship between expression these genes with clinicopathological and prognostic parameters.Results:The genome-wide expression profiling showed that expression of1326genes was increased by at leased2-fold in cervical cancer over that in normal cervix,while expression of1432genes was decreased by at least2-fold in cervical cancer over that in normal cervix.7in10of the apoptotic pathways were over-expressed in cervical cancer.The anti-apoptotic genes c-IAP1of the IAP family and Bcl-xl of the Bcl2family were expressed at high levels in cervical cancer,while the anti-apoptotic gene Bcl2was expressed at low level in cervical cancer. Validation of microarray data was done at both mRNA and protein levels,which further demonstrated the following Results:The mRNA of anti-apoptotic genes c-IAPl,Bcl-xl were expressed at high levels and Bcl2were expressed at low levels in cervical cancers,The protein expression level of anti-apoptotic genes c-IAPl,Bcl-xl were high and Bcl2was low in cervical cancers.Conclusion:(1)proving the occurrence of invasive cervical cancer is a multistep genetic pathways regulated more complex process of biological molecular marker spectrum of cervical cancer and screening of tumor marker gene, to study the molecular mechanism of cervical cancer occur in xinjiang and the early clinical diagnosis and treatment of bladder cancer has important significance.lt is helpful for us to get further understanding on the molecular mechanism of development and signal transduction pathway for study of common differentially expressed genes between them.(2)cervical cancer tissue of IAP family and Bcl2family anti-apoptotic proteins high expression may play an important role in cervical cancer anti-apoptotic mechanisms, these genes can be used for cervical cancer susceptibility prediction not only early diagnosis and disease monitoring, for the the development of molecular targeted therapy also has important guiding significance.
