Objective:To study the effect tranexamic acid on cell proliferation and melanin synthesis of normal human melanocytes. Further work to explore the mechanism underling restrain differention action by tranexamic acid, the study was performed to investigate whether tranexamic acid effect the mRNA of tyrosinase、tyrosinase related protein1and microphthalmia associated transcription factor. Methods:Melanocytes of prepuces of healthy men were cultured and logarithmic growth phase cells were choosed to develop.After ultraviolet irradiation the melanin cells were cultured with different concentrations of tranexamic acid, and the blank was referenced.After incubation24-72hours, cell proliferation was measured with cck-8assay;The synthesis of melanin was measured with NaOH method;The tyrosinase activity was detected by DOPA-oxidation;Semiquanttitative reverse transcriptase polymerase chain reaction was performed to assess the mRNA expression levels of tyrosinase、tyrosinase related protein-1and microphthalmia associated transcription factor. Results:Tranexamic acid couldn’t suppress the proliferation of cultured melanocyte with every concentration compared with the untreated control(P>0.05).Tranexamic acid could signifieantly suppress melanin sythesis at1000mg/ml(P<0.05). Tranexamic acid with500mg/ml and1000mg/ml suppressed tyrosinase activity respectively(P<0.05).The effect of tranexamic acid on melanocyte was dosed dpended. Microphthalmia associated transcription factor mRNA level restrained in melanocyte treated with1000mg/ml concentration compared with untreated control(P<0.05).Tyrosinase mRNA level and tyrosinase related protein-1mRNA level restrained respectively in melanocytes treated with500mg/ml and1000mg/ml concentration compared with untreated control(P<0.05). The effect of tranexamic acid on mRNA was dosed dpended(P<0.05).Conclusion:Tranexamic acid could suppress the tyrosinase activity and melamin synthesis of melanocytes. But tranexamic acid couldn’t suppress proliferation of melanocyte. Moreover tranexamic acid could suppress mRNA levels of microphthalmia associated transcription factor and tyrosinase、tyrosinase related protein-1,which thereby leads to decreases in melanin producted by melanocytes.
