PART ONE:The pathogenesis of reflux esophagitis associated with lysolecithin.(一) Effect of lysolecithin and Hydrochloric acid on the morphological changes and the content of IL-8、PGE2 of rats’esophageal tissue in reflux esophagitis model.Objective:To explore the effect of lysolecithin and Hydrochloric acid on the morphological changes and the content of IL-8 and PGE2 of esophageal tissue in reflux esophagitis model. Method:A total of 40 healthy SD rats were randomly divided into NS control group(n=20), model group(n=20), NS control group and model group were divided into two subgroups(7d group、14d group) according to different perfusion time. NS control group and model group were administered with diluted NS、lysolecithin with Hydrochloric acid respectively in the esophagus for 7days、14days. Then rats were sacrificed at 7day、14day and the esophagus was obtained, the morphological changes and the content of IL-8、PGE2 was examined by optical microscope and radioimmunoassay respectively.Results:1.At the end of preparation of model, morphological changes including inflammatory cell infiltration, vacuolar degeneration of epithelial cell; erosion even ulceration was seerr in model group.2. At the end of preparation of model, IL-8 and PGE2 content as well as the index of mucosa injury of model group was significantly high than that of NS control group.3. IL-8 and PGE2 content as well as the index of mucosa injury of model group increase with the lengthen of perfusion time. Conclusions:Lysolecithin mixed with Hydrochloric acid can increase the level of IL-8 and PGE2 of rats’ esophageal tissue. The occurence and development of esophagitis associated with lysolecithin mixed with Hydrochloric acid may be related to the level of IL-8 and PGE2.PART ONE:The pathogenesis of reflux esophagitis associated with lysolecithin.(二)Effect of lysolecithin and Hydrochloric acid on the morphological changes and the content of GAS、MTL、VIP of rats’esophageal tissue in reflux esophagitis model.Objective:To explore the effect of lysolecithin and Hydrochloric acid on the morphological changes and the content of GAS、MTL、VIP of esophageal tissue in reflux esophagitis model. Method:A total of 60 healthy SD rats were randomly divided into NS control group(n=30), model group(n=30), NS control group and model group were divided into three subgroups(10d group、14d group、21 d group, n=10) according to different perfusion time. NS control group and model group were administered with diluted NS、lysolecithin with Hydrochloric acid respectively in the esophagus for 10 days、14day、21 days, Then rats were sacrificed at 10 days 、14day、21 days. The esophagus was obtained, the morphological change was examined by optical microscope and the content of GAS、MTL、VIP of serum was detected by radioimmunoassay respectively. Results:At the end of preparation of model, morphological changes including inflammatory cell infiltration, vacuolar degeneration of epithelial cell, erosion even ulceration was seen in model group. At the end of preparation of model, the content of VIP of serum as well as the index of damage injury of model group was significantly high than that of NS control group. The content of GAS、MTL of serum in model group was significantly lower than that of control group. Conclusions:Lysolecithin mixed with Hydrochloric acid can decrease the level of GAS、MTL of rats’serum, and increase the level of VIP, The occurence and development of esophagitis associated with lysolecithin mixed with Hydrochloric acid may be related to the level of GAS、MTL、VIP of serum. The clearance properties is lessened and the reflux of gastric contents is increased. The inflammation reaction is weightened and the lower esophageal sphincter relaxation is increased. Thus esophagitis associated with lysolecithin mixed with Hydrochloric acid occurs.PART TWO:Effect of Kangfuxin on the morphological changes and the content of IL-8 and PGE2 of rats’esophageal tissue in reflux esophagitis model and the possible protection mechanismObjective:To explore the effect of Kangfuxin on the morphological changes and the content of IL-8 and PGE2 of esophageal tissue in reflux esophagitis model and the possible protection mechanism. Method:A total of 60 healthy SD rats were randomly divided into Aluminum phosphate gel group (n=10), Kangfuxin group(n=10),model group(n=20),control group(n=20),all groups except control group were administered with diluted lysolecithin with Hydrochloric acid in the esophagus for 14 days. Then ten rats of model group and ten rats of control group were sacrificed. The esophagus was obtained, the morphological changes and the content of IL-8、PGE2 was examined by optical microscope, electron microscope and radioimmunoassay respectively. The next day model group was given 0.9%NaCl, while treatment groups were given Aluminum phosphate gel, Kangfuxin respectively for another 14 days. Control group was given nothing. Afterwards the rats were sacrificed and the above detections were accomplished in the same way. Results:At the end of preparation of model, morphological changes including inflammatory cell infiltration, vacuolar degeneration of epithelial cell, erosion even ulceration was seen in model group. Ultrastructural changes showed severe detachment of epithelial cells. While control group shows normal mucosa. At the end of treatment, IL-8 and PGE2 content as well as the index of mucosa injury of each treatment group was significantly lower than that of model group. There are no significantly difference on IL-8 and PGE2 content as well as the index of mucosa injury between Aluminum phosphate gel group and Kangfuxin group.Conclusions:Kangfuxin is effective in the treatment for reflux esophagitis induced by lysolecithin mixed with Hydrochloric acid, The possible protection mechanism may be related to reducing the level of IL-8 and PGE2.PART THREE:Effect of Simotan on the morphological changes and the content of GAS、MTL、VIP of rats’ serum in reflux esophagitis model and the possible mechanismObjective:To explore the effect of Simotan on the morphological changes and the content of GAS、MTL、VIP of rats’serum in reflux esophagitis model and the possible mechanism. Method:A total of 40 healthy SD rats were randomly divided into normal group (n=10), NS group (n=10), simotan group (n=10), mosapride group(n=10). Except normal group, each group was administered with diluted lysolecithin with Hydrochloric acid in the esophagus for 14 days. The next day NS group were given 0.9%NaCl, while treatment groups were given simotan、mosapride respectively for another 10 days. Afterwards the rats were sacrificed. The esophagus was obtained, the morphological changes was examined by optical microscope, and the content of GAS、MTL、VIP in serum was dectected by radioimmunoassay. Results: Morphological changes including inflammatory cell infiltration, vacuolar degeneration of epithelial cell, erosion even ulceration was seen in model group, while normal group shows normal mucosa. The index of mucosa of simotan groups is lower than that of NS group(P< 0.05), The content of GAS、MTL of simotan groups is higher than that of NS group(P<0.05). The content of VIP of simotan groups is lower than that of NS group(P<0.05). Above indicators between Simotan group and mosapride group were no difference(P>0.05).Conclusions:Simotan is effective in the treatment for reflux esophagitis induced by lysolecithin mixed with Hydrochloric acid. The possible protection mechanism may be related to reducing the level of VIP and increase the level of GAS、MTL.
