| ObjectiveAnalyses how allergic asthma affects the lungs, Immune proteins, cell factor of cavy and observes how acupoint catgut embedding affects the lungs, Immune proteins, cell factor of allergic asthma cavy model, so we can understand the mechanism and affect of allergic asthma and then promote the cure of it. Researching the mechanism of acupoint catgut embedding curing allergic asthma, to provide new ideas and methods for curing allergic asthma and find a better therapy.Methods40 healthy cavies, half male, half female, weighted about 230-280g, were kept in quiet environment for 2 days. Divided the cavies into control group, model group, acupoint catgut embedding group and Dexamethasone group,10 cavies each. Established allergic asthma model by lml 10% egg protein solution Intraperitoneal injection at the 1st day, and aerosol inhalation spray inhalation of 1% egg protein solution from the 15th day,20minutes every day and lasting for 2 weeks. As cavies began to buck, move restlessly, breathe rapidly and intensified, Abdominal muscle spasm, we built the allergic asthma cavy model successfully. Control group wouldn’t do anything until the 30th day, and then lavaged with Distilled water,2ml each every day. Model group, kept being allergic for 2 weeks, have aerosol inhalation spray inhalation of 1% egg protein solution from the 15th day to trigger asthma,20minutes every day and lasting for 2 weeks. Acupoint catgut embedding, began the therapy at the 15th day, before aerosol inhalation spray inhalation every day. located the acupoint of "BL11", "BL12" and "BL13", embeded the catgut with 7# syringe needle by 5cm beneath the skin and pushed the catgut into the acupoint with Acupuncture needle. Once a week, and changed to another side each time. Dexamethasone group, did the Intraperitoneal injectiong with 0.5mg/kg Dexamethasone 30minutes before aerosol inhalation spray inhalation. At last, kill all the cavies and have the Immunohistochemistry detection.Results1.Lung tissue:Ccontrol group has a complete bronchial epithelium integrity and normal lung tissue. In model group we can see a large number of inflammatory cell, eosinophil (Eos), neutrophil (Neu) and lymphocyte (Lym) as main, around bronchial epithelium and vesse. We can see a large number of inflammatory cell at pulmonary interstitium. Submucosal is thickened. Mucosal fold is increased. Lumen of bronchial epithelium is narrowered. After the intervention of Dexamethasone and acupoint catgut embedding, the change mentioned above is alleviated.2. Inflammatory cell:Comparing model group, acupoint catgut embedding group and Dexamethasone group with control group at the number of inflammatory cell, the difference is statistically significant (P<0.05). Comparing acupoint catgut embedding group and Dexamethasone group with model group, the difference is statistically significant (P<0.05). Eos has the The most minority number in the three of inflammatory cell in acupoint catgut embedding group and Dexamethasone group, the difference is statistically significant (compared with Lym and Neu, bothP<0.05). Eos has the The maximum number in the three of inflammatory cell in model group, the difference is statistically significant (compared with Lym and Neu, both P<0.05). Acupoint catgut embedding group and Dexamethasone group can reduce inflammatory cell, Eos as main, in lung tissue.The difference is statistically significant (P<0.05).Allergic asthma lung tissue contain a large number of inflammatory cell, Eos as main. Acupoint catgut embedding and Dexamethasone can alleviate this case.3. Immune proteinsis:Model group has a higher average optical and Positive rate of IL-21 and IL-21R than control group. The difference is statistically significant (P<0.001).Comparing Dexamethasone group and acupoint catgut embedding with model group, average optical and Positive rate of IL-21 and IL-21R are much lower, The difference is statistically significant (P<0.01)。Comparing and acupoint catgut embedding with Dexamethasone group, The difference is statistically significant (P<0.01) Both acupoint catgut embedding and Dexamethasone can reduce average optical and Positive rate of IL-21 and IL-21R but acupoint catgut embedding is not as good as Dexamethasone.4. Cell factor:Model group has a higher average optical and Positive rate of ICAM-1 and VCAM-1 than control group. The difference is statistically significant (P<0.001).Comparing Dexamethasone group and acupoint catgut embedding with model group, average optical and Positive rate of ICAM-1 and VCAM-1 are much lower, The difference is statistically significant (P<0.01)。 Comparing and acupoint catgut embedding with Dexamethasone group, The difference is statistically significant (P<0.01). Both acupoint catgut embedding and Dexamethasone can reduce average optical and Positive rate of ICAM-1 and VCAM-1 but acupoint catgut embedding is not as good as Dexamethasone.Conclusion1. Allergic asthma can affect the lung tissue of cavies, inducing inflammatory. Comes cellular infiltration. Submucosal is thickened. Mucosal fold is increased. Lumen of bronchial epithelium is narrowered. Acupoint catgut embedding and Dexamethasone can alleviate such affection and alleviate allergic asthma.2. Eos is the main inflammatory cell in cellular infiltration induced by allergic asthma. Acupoint catgut embedding and Dexamethasone can reduce the number of Eos equivalently. Both acupoint catgut embedding and Dexamethasone can cure allergic asthma.3.
