ObjectiveIn order to discuss the cerebral nerve protective mechanism of Jin three needle pretreatment on Ischemia reperfusion injury in rats, this topic tests brain tissue heat shock protein 70 (HSP70), IL-1, IL-6 and TNF-protein expression and serum Pro adrenal cortical hormone (ACTH), by acupuncture of the temporal three-needle and Baihui.MethodOn this research aspect,32 rats, SPF grade, were randomly divided into sham operation group, nature recovery group, acupuncture treatment group and Nimodipine group with a scale of 1:1:1:1. The sham operation group:Simulated surgery to separate the internal carotid, external arteries, do not carry out the next surgery, suture tissue and skin. Nature recovery group:Do nothing after the cerebral ischemia reperfusion, but give a stimulus every day. Acupuncture group:By acupuncture of the temporal three-needle and Baihui once a day,15 minutes each time, give a strong stimulation for 1 minute at the beginning and end,5 days a course. Nimodipine group:3 times of adult dose is given by stomach,5 days a course. Using a modified Bederson 5 grades, to animals of neurological symptoms grading evaluation, rats were sacrificed at the end of the experiment, determination of the content of serum ACTH by radioimmunoassay, immunohistochemical detection of chemical of HSP70 in brain tissue of rats, IL-1, IL-6 and TNF-protein expression.Results1. Rats of model group, acupuncture group, Nimodipine group, the neural function score were increased, and model group increased most significantly, the acupuncture group of neural function defect score was significantly lower than that of model group. The statistical difference is significant (P< 0.05),Rats of Nimodipine group, the neurological function scores and the acupuncture group compared by statistical difference is significant (P< 0.05).2. Rats of model group, acupuncture group, Nimodipine group, the ACTH were increased, and model group increased most significantly, compared with the model group, acupuncture group, Nimodipine group experimental rat serum ACTH content were significantly decreased, and the difference is significant (P<0.05), Nimodipine group of experimental rats serum ACTH content and acupuncture group compared by statistical difference no significant (P> 0.05;.3. Rats of model group, acupuncture group, Nimodipine group, the cerebral PSP70 protein expression compared with the sham operation group increased. The model group increased most significantly, the acupuncture group, nylon membrane in the same group experimental rat cerebral HSP70 protein expression were lower than those in the model group and the statistical difference is significant (P<0.05), nylon membrane in the same group experimental rat cerebral HSP70 protein expression and acupuncture group compared by statistical difference is significant (P<0.05).4. Model group and the acupuncture group experimental rat brain tissue IL-1 beta and IL-6 protein expression compared with the sham operation group were different degree increased, and model group increased most significantly, the acupuncture group, nylon membrane in the same group experimental rat brain tissue IL-1 beta and IL-6 protein expression were lower than those in the model group and the statistical difference is significant (P<0.05), nylon membrane in the same group experimental rat brain tissue IL-1 beta and IL-6 protein expression and acupuncture group compared by statistical difference is significant (P<0.05).5. The model group, the acupuncture group and the experimental rat brain tissue TNF-protein expression compared with the sham operation group were different degree increased, and model group increased most significantly, the acupuncture group, nylon membrane in the same group experimental rat brain tissue TNF-protein expression were lower than those in the model group and the statistical difference is significant (P<0.05), nylon membrane in the same group experimental rat brain tissue TNF-protein expression and acupuncture group compared by statistical difference is significant (P< 0.05).Conclusion1. Jin three-needle can reduce Ischemia reperfusion injury neural function defect score effectively and improve the ability of learning and memory of rats in the experiment.2. Jin Three-needle can effectively reduce the HSP70 in brain tissue of rats, IL-13, IL-6 and TNF-α, decreased serum ACTH release produced protective effects on the brain, which may be the main mechanism.
