Objective1、To prepare a new type of perfluoropentane-loaded hollow magnetic microsphere (PFP-HMPMS). To investigate the basic character of this elaborate designed microsphere, the possibility of its phase change promoted by different temperature cutoffs, and its impact on the result of MWA.2. To investigate the capacity of PFP-HMPMS engulfed by HepG2 cell line, and the cytotoxicity of PFP-HMPMS. To investigate the feasibility of PFP-HMPMS enhanced the therapeutic effects of MWA for the HepG2 liver cancer cells in vitro.3. To determine the feasibility of PFP-HMPMS enhanced the efficiency of MWA in nude mice transplanted subcutaneous HepG2 tumor in vivo.Materials and Methods1. The HMPMS was prepared using template method and its appearance and size was analyzed. The possibility of phase change of PFP-HMPMS promoted by different temperature and its impact on the result of MWA were analyzed.2. Perls stain was performed to evaluate the phagocytosis of PFP-HMPMS in HepG2 cell line. MTT method was used to masure the influence of PFP-HMPMS on the proliferation of HepG2 cells. Flow cytometry was adopted to compare the proportion of cell state after MWA at different temperature with or without swallowing of PFP-HMPMS.3. Transplanted subcutaneous HepG2 tumor in nude mice was established and MWA was performed on each tumor-bearing mice. The rate of temperature rising and histopathological changes were compared between the two groups with or without local injection of PFP-HMPMS.Results1. The elaborate designed HMPMS was with good sphericity and uniform particle size. Temperatue higher than 40℃ could promote a phase transition of the PFP-HMPMS. The HMPMS may accelerate the temperature rising rate of MWA.2. The HMPMS can be engulfed by HepG2 cell and the positive labeled rate was 71.7《》3.5%. PFP-HMPMS with a concentration of 100μg/ml may slightly restrain the proliferation of HepG2 cells. Flow cytometry showed that the percentage of normal live and early apoptosis cells were significantly increased in the PFP-HMPMS group at the MWA heating temperature cut-off of 45℃,50℃ and 60℃ respectively. The percentage of early apoptosis and late apoptosis or dead cells were significantly increased in the PFP-HMPMS group at the temperature cut-off of 50℃ and 60℃3. After local injection of PFP-HMPMS, the required time for temperature rising to 50℃ shortened obviously compared to the control group. Histopathological results showed that the rumor structure changed in the microsphere deposition area.Conclusion1. The prepared FPF-HMPMS can reach a liquid-gas phase transition through a temperature rise. The HMPMS may accelerate the temperature rising rate of MWA.2. The HMPMS can be engulfed by HepG2 cell and has slight influence on cell proliferation. Under the condition of same temperature control, FPF-HMPMS can increase the proportion of tumor cell apoptosis and death.3. FPF-HMPMS can improve the efficiency of temperature rising of MWA, and enhance the effect of MWA in the treatment of tumors in vivo.
