Objective: Based on the Black Dihuang Pill in good clinical treatment of chronicrenal failure, this article from the perspective of the experimentation observes blackDihuang Pill for5/6nephrectomy to chronic renal failure rat renal function, urinary protein,oxidative stress, pathological morphology, the expression of cytokines and cell apoptosis,and this article explores the black pill resistance mechanisms of renal fibrosis observationto provide new ideas for the clinical treatment of renal fibrosis prevention.Methods:1. Setting up5/6nephrectomy in the rat model with chronic renal failure;2.Detecting serum creatinine, urea nitrogen, and24hours urinary protein content changes;3.The enzyme standard method to detect renal tissue and serum MDA, SOD content;4.Using HE staining to observe the renal tissue pathological changes;5.Using theimmunohistochemical method to test organization TGF-β1, the exPression of CTGF andPDGF-BB;6. RT-PCR method is used to detect kidney cell apoptosis.Results:(1) The Black Dihuang Pill of high, medium and low dose groups, Niao DuQing group as well as los Ding Xin group can improve the rat kidney, resist oxidativestress, reduce urinary protein, reduce the glomerular sclerosis index and the fibrosis indexof renal tubule, reduce the expression of TGF-β1, CTGF, PDGF-BB semi-quantitative,and inhibit cell apoptosis, and it has a very significant difference (P <0.01) compared withmodel group.(2) Black pill effect of high dose group sets a basic quite function with NiaoDu Qing group and is better than los Ding Xin (P <0.05)in improving renal function,antioxidation, cutting BAX, Caspase3, and raising the expression of BCL-2;(3) LosDing Xin effect is better than the Black Dihuang pill group and Niao Du Qing group inreducing urinary protein (P <0.05). Conclusion: BlackI Dihuang pill high dose group has the functions of reducing ratsserum BUN, SCr levels, reducing proteinuria in rats, improving the nutritional status ofrats; reducing the kidney tissues and serum MDA level, improving the level of kidneytissues and serum SOD, and has a significant antioxidant effect; Black Dihuang pill highdose group can reduce the glomerular sclerosis index and the fibrosis index of renal tubuleand inhibite the excessive expression of TGF-β1, CTGF, PDGF-BB in the renal tissue;Black pill high dose group can cut the expression of Bax, Caspase-3, Caspase-8,Caspase-9,and improve the expression of BCL-2, inhibiting cell apoptosis. For the reasons above theBlack Dihuang pill high dose group has the effect of prevention and treatment of renalfibrosis.
