Objective: On the basis of current domestic and international Chinese herbal system of quality assessment methods, select Shandong authentic ingredients S corpion as samples, establishes biological fingerprints, chemical fingerprints of t he active ingredient of Scorpion. Evaluate the quality of Chinese medicine at v arious aspects from the qualitative and quantitative scorpion species identificatio n and genetic material to identify the chemical composition, and explore a new formulations of Scorpion for future research.Method: Established the biological fingerprints using protein polyacrylamide gel electrophoresis(PAGE), random amplified polymorphic DNA(RAPD). Esta blished the chemical constituents fingerprints using ultraviolet spectroscopy(U V), High performance liquid chromatography(HPLC) and gas chromatographymass spectrometry(GC-MS). Combined the findings of this study and the 2010 version of Chinese Pharmacopoeia, fomulated the quality standard(draft) and the drafting of instructions of Scorpion in Shandong province, and establishmen t of quality evaluation system Scorpion.Results:1. The UV spectra of four kinds of extraction solvent of SCORPION were established. The results showed that UV spectra of SCORPION from different regions were very close, had the common characteristic peaks, showed that the main component was the same or similar.2. The HPLC fingerprint with Phillyrin as internal standard reference peak o f SCORPION was established. 19 common characteristic peaks were determined,and the common peaks area accounted for more than 90% of the total area.3. Chemical compositions of the fat-soluble ingredients from SCORPION wer e determined by GC-MS analysis. 9 common peaks were determined, and the common peaks area accounted for more than 90% of the total area.4. The SCORPION seed PAGE fingerprint was set up.6 common bands were determined, and the color depth, width, mobility was similar. The common pe aks area accounted for 79.2% to 97.9% of the total area.5. The results of RAPD analysis showed that polymorphic bands percentage was 73.91%, showed genetic diversity.6. Fomulated the quality standard(draft) and the drafting of instructions of Scorpion in Shandong province, and establishment of quality evaluation system Scorpion.Conclusion: In this study, the quality of research was carried out by more c omprehensive and systematic on Shandong authentic ingredients SCORPION. M aked a research on different origin Scorpion UV, HPLC fingerprint, Scorpion f at-soluble fractions and proportional relationship, biological fingerprint. Initially established quality evaluation system of Scorpion in Shandong province and pr ovided a more accurate and effective evaluation methods and systems basis of identifing and evaluating the quality of scorpion species。...
