Effect Of Acupuncture Preconditioning At Jiaji Points On The Expression Of Inflammatory Cytokines Of Myocardial Ischemia Reperfusion Injury

Objective: To observe the protective function of Acupuncture pretreatment at Jiaji(EX-B2) on the cardiac function of rats with myocardial ischemia reperfusion injury(MIRI), and expression of inflammatory cytokines of cardiomyocytes, and explore the anti-inflammatory mechanism of acupuncture pretreatment on myocardial ischemia reperfusion injury.Methods: 50 rats were divided into 5 groups: a sham-operation-group(control group), a model group, a Jiaji group, a Neiguan group, and a Yangliangquan group. Ten rats for each group. The acupuncture groups were all pretreated for 7 days, and then use the model group and acupuncture groups to copy MIRI models with the method of ligating the anterior descending coronary artery. Measure the TNF-α and IL-1β content in serum and ischemic myocardium in each group by enzyme-linked immunosorbent assay and measure the content of serum cTnI. Determine expression of TNF-α, IL-1β, and IFNγ gene in ischemic myocardium tissue by real-time fluorescence quantitative PCR method.Results: 1. Compared with that in the sham-operation-group, in model group, level of inflammatory cytokines in serum, including cTnI and IL-1β, increased(P <0.05), and the inflammatory cytokines in ischemic myocardium tissue, including TNF-α, IL-1β, and IFNγ genes, were up-regulated(P <0.05).2. Compared with that in the model group, in Jiaji group, content of serum cTnI and IL-1β decreased(P<0.05), TNF-α, IL-1β and IFNγ genes in ischemic myocardium tissue were down-regulated(P <0.05); in Neiguan group, content of serum cTnI、IL-1β decreased(P<0.05), IL-1β and IFNγ genes in ischemic myocardium tissue were down-regulated(P <0.05) while TNF-α in ischemic myocardium tissue was up-regulated(P <0.05).3. Compared with that in Yanglingquan group, in Jiaji group, content of serum cTnI and IL-1β decreased(P<0.05), TNF-α, IL-1β and IFNγ genes in ischemic myocardium tissue were down-regulated(P <0.05).4. Compared with that in Neiguan group, in Jiaji group, content of serum IL-1β decreased(P<0.05), TNF-α gene in ischemic myocardium tissue were down-regulated(P <0.05) while IL-1β gene in ischemic myocardium tissue were up-regulated(P <0.05) and the expression difference of TNF-α gene in serum and IL-1β gene in ischemic myocardium was not statistically significant.Conclusion: Jiaji pretreatment has a significant protective effect on myocardial ischemia-reperfusion injury. The mechanism may be associated with the decrease of the content of TNF-α, IL-1β and IFNγ in ischemic myocardium of rats with myocardial ischemia reperfusion injury.
