ObjectiveTo investigate the intervention effect of prescription of Dahuang zhechong Pill on Lewis lung cancer tumor bearing mice passive smoking under the condition of immune status (CD4+D25+Foxp3+level and the expression of Foxp3 gene expression), provide theoretical and experimental basis for improvement and prevention of lung cancer patients’quality of life.Method1. by Lewis lung cancer tumor bearing mice:Kunming mice of clean grade selection, the cultured mouse Lewis lung cancer cells (LLC), diluted with physiological saline, adjust the cell concentration is 1×107. In the mouse right axilla subcutaneous injection of 0.2ml only modeling. After 10 days, the axillary into 64 tumor 0.5cm* 0.6cm above mice were randomly divided into three groups:model control group (model group), passive smoking group, Dahuang zhechong pill group cisplat in group, did not cause the mouse model as blank control group (blank group), each group has 16, a total of 5 groups.2. Administration:passive smoking group, Dahuang zhechong Pill group and cisplatin group mice were treated with passive smoking, namely in moving type exposure cabinet, take dynamic inhalation of cigarette smoke, mainstream smoke and side stream smoke control in 11% and 89% respectively. Daily exposure to 6h. At the same time, Dahuang zhechong Pill group were given Dahuang zhechong pill administration by 1.37g/kg/d, gastric lavage volume 0.4ml,1 times a day, continuous 28d. The cisplatin group was treated with cisplatin by lmg/kg intraperitoneal injection (iP), fourth,11,18, 25 days each 1 times. The blank group and model group were perfused with equal volume of distilled water,0.4ml,1 times a day,28d.3. the detection index:on 7d,14d,21d and 28 d, were observed in the spirit, activities, hair color, diet, respectively on 14 and 28 days measured mice weight, spleen index, thymus index, tumor inhibition rate; detecting the content of IL-2 in serum, spleen CD4 +CD25+Foxp3+Treg accounted for CD4+T cell and the ratio of transcription factor Foxp3 gene expression; each mouse lung tissue was embedded in paraffin, he staining, light microscope observation mice lung tissue morphological change.Result1. General conditions:with the blank group, model was made in mice after spirit, activity, fur, food intake and water intake, body weight growth were affected Increase the amount (P< 0.05); including passive smoking group of the most obvious, weight growth slowed down significantly (2 weeks P<0.05,4 weeks P< 0.01); and passive smoking group, Dahuang zhechong Pill group spirit has improved activity and food intake increased, weight growth Jiao Mingxian (P<0.01) and cisplatin group spirit, and no obvious change, body weight decreased significantly (2 weeks P<0.05,4 weeksP<0.01) and cisplatin group, Dahuang zhechong Pill group mice mental state improved, activity and diet, weight increased significantly (P<0.05).2. Organ index:compared with the control group, after the establishment of the model mouse organ index were decreased, the cisplatin group decreased the most obvious,2 weeks spleen index have significant difference (P<0.05),4 weeks thymus index and spleen index decreased very significant difference (P<0.01); and passive cigarette smoking group, Dahuang zhechong Pill group internal organs index improved,2 weeks, there was no significant difference,4 weeks when there were significant differences (P< 0.05); compared with the cisplatin group, improve the Dahuang zhechong Pill group of mice spleen index and significant difference (P<0.05).3. Morphological change:compared with the control group, after modeling in lung tissues of mice with visible pleomorphic cells, the cell nucleus was big and deep staining, visible abnormal mitosis; passive smoking can make the mouse lung pleomorphic cells increased significantly, the visible pleomorphic cells into semi annular and circular permutation, formed around the lesions associated with emphysema; Dahuang zhechong Pill reduce passive smoking in the lung tissue of the mice pleomorphic cells, alleviate lesion; cisplatin in lung tissues of mice with pleomorphic cells decreased significantly, alleviate lesion, individually visible pleomorphic cells, occasionally nuclear division.4 weeks after modeling mice shaped cells compared with two weeks more, visible cellular atypia into semi annular and circular permutation and alveolar septal thickening and inflammatory cell infiltration phenomenon.4. The immune indexes of IL-2:compared with the control group, serum IL-2 content was decreased after modeling, the mice in model group have significant difference (P<0.05 for 2 weeks,4 weeks P <0.01), which passive smoking group decreased most significantly, have very significant difference (P<0.01); and passive smoking group, Dahuang zhechong Pill group, cisplatin group, serum IL-2 content were increased, the differences were statistically significant (P<0.05); Dahuang zhechong Pill group and cisplatin group compared Dahuang zhechong Pill group improve obviously, but no significant difference (P>0.05).5. immune index CD4+CD25+Foxp3+:2 weeks, compared with the blank control group, mice model of spleen CD4+CD25+Foxp3+Treg accounted for CD4+T cell percentage increased, the passive smoking group increased most significantly, and there is a significant difference (P<0.01); compared with the passive smoking group, Dahuang zhe chong pill group, cisplatin group mouse spleen tissue of CD4+CD25+Foxp3+Treg accounted for the proportion of CD4+T cells decreased, cisplatin group decreased obviously, but there were no significant difference (P>0.05). Compared with the cisplatin group, Dahuang zhechong Pill group increased, but there was no significant difference (P>0.05).After 4 weeks, compared with the blank control group, mice model of spleen CD4+CD25+Foxp3+Treg accounted for CD4+T cell ratio increased, the differences were statistically significant (P< 0.05); compared with the model group, passive smoking group, Dahuang zhechong Pill group, cisplatin group mice spleen CD4+CD25+Foxp3+Treg accounted for the proportion of CD4+T cells were decreased. There was no significant difference (P>0.05); compared with the passive smoking group, Dahuang zhechong Pill group, cisplatin group were down regulated, cisplatin group decreased obviously, but there were no significant difference (P>0.05).6. Foxp3 gene expression:after 2 weeks, compared with the blank group, made mice model of spleen tissue of Foxp3 gene expression was enhanced, passive smoking group is most obvious, have very significant difference (P<0.01); compared with the passive smoking group, Dahuang zhechong Pill group, cisplatin group spleen tissue of Foxp3 gene table was decreased, and there was a significant difference (P<0.01); Dahuang zhechong Pill group and cisplatin group compared to cisplatin group spleen tissue of Foxp3 gene expression decreased, there was significant difference (P<0.05).After 4 weeks, compared with the blank group, model group, Dahuang Zhechong Pill group and cisplatin group mice Foxp3 gene expression levels were increased in model group, there was significant difference (P<0.01), passive smoking group of mice Foxp3 gene expression level decreased, there was significant difference (P<0.05); compared with the model group, passive smoking group, Dahuang zhechong Pill group and cisplatin group were decreased, passive smoking group, cisplatin group showed significant difference (P<0.01), no difference between the group of Dahuang zhechong pill (P>0.05); compared with the passive smoking group, Dahuang zhechong Pill group and DDP group were increased, there were significant differences (P<0.05); Dahuang zhechong Pill group and cisplatin group of mice Foxp3 gene expression levels had no significant difference.Conclusion1. Lewis lung cancer cells can cause the formation of the tumor block in the mice of Kunming.2.Passive smoking has a significant effect on the general situation, weight, organ index, morphological changes and immune index of Lewis lung cancer mice (P<0.05 or P<0.01).3. Dahuang zhechong Pill on Lewis lung cancer tumor bearing mice passive smoking conditions generally, weight, organ index, morphology changes and immune indexes were significantly improved and immune regulation effect (P<0.05 or P<0.01), and cisplatin group, there is no significant difference. The general situation, weight and organ index changes were better than that of cisplatin group (P<0.05).4. Dahuang zhechong Pill of tumor bearing mice with Lewis lung cancer in general, body weight, organ index is superior to cisplatin group, may is by increasing the serum IL-2 content, reduce mouse spleen tissue of CD4+CD25+Foxp3+Treg accounted for CD4+T cell and the ratio of Foxp3 gene expression...
