Object:CGRP is very important in protecting the cardiac muscle. Chinese medicine is our Chinese quintessence, which has been regard as a medicine to resist fatigue. This research intends to make the model of hard training rats, and observing changes of indexes, CGRP and CGRP mRNA, after6weeks training and intake Chinese medicine. Through this experiment, we want to find out the mechanics behind the Chinese medicine which could protect cardiac muscles.Method:50eight-week-old male SD rats. Rates are separated into5groups, with10for each group (N=10). There are quiet group with no medicine (A), the quiet and fed medicine group (B), exercise group (D), exercise with medicine group (E), exercise and the high-dose group (F group). Rates were fed normally, group B and group E rats were digested by0.1g/kg traditional Chinese medicine, group F with0.2g/kg traditional Chinese medicine concentration, other group fed the same amount saline. All groups had been trained for6weeks. After that, rates were killed immediately with peritoneal anesthesia. Experiment took blood and heart to be samples. Experiment used colorimetric determination of serum antioxidant enzymes SOD, GSH-Px content radioimmunoassay Determination of serum and myocardial tissue calcitonin gene-related peptide (CGRP) content, using RT-PCR detection of myocardial tissue content of CGRP mRNA.Result:1. Comparing to group A, Group D has significant increase of serum ALT, LDH, and a-HBDH. Comparing to Group D, Group E has significant decrease of serum ALT, LDH, CK, and a-HBDH. Comparing to group E, group F has significant increase of serum LDH, with no significant changes among other indexes.2. There are no significant changes in serum SOD, and GSH-Px in each group.3. Changes of CGRP:Comparing to Group A, Group D has increasing tide, without any significant reluctance. But the myocardial tissue has decreased significantly. Comparing to GroupD, Group E has significant increasing of serum CGRP, without obvious changes in myocardial tissue. But Group F has significant increasing of serum CGRP, with significant decreasing in myocardial tissue. There are no significant changes between group E and F.4. Expression of α-CGRP mRNA: Comparing to Group A, group B has no significant changes, but group D has significant increasing. Comparing group D, group E has significant decreasing, and group F also has significant decreasing.Conclusion:1. After6weeks incremental load training, there more injuries happened among cardiac muscle. Simultaneously, CGRP’s contents has been decreased with much expression of CGRP mRNA.2. Chinese medicine can effective inhibit ascending of serum cardiac marker enzyme activity and alleviate cardiac injured caused by high seep of hear rat. Through enhancing CGRP’s content in serum, cardiac injury will be mitigated. However there is no distinguishing difference between two dosages.
