| ObjectiveBRCA1, TYMS, TUBB3, TOP2A, PTEN are closely related with the chemotherapy of breast cancer. In this study, we used the liquid-phase chip to detect the expression of BRCA1, TYMS, TUBB3, TOP2A, PTEN genes’mRNA, found the relevance with the clinical and pathological indicators, analyzed the relationship with neoadjuvant chemotherapy, provided the basis for individualized treatment of breast cancer patients.Method794patients with phase II A to IIIC breast cancer received neoadjuvant chemotherapy treatment of TAC/TEC/PAC/PEC scheme for four cycles, divided into548responders and246unresponders. Chose23responders into Group A and23unresponders into Group B by random. Take a biopsy specimens detect the expression of BRCA1, TYMS, TUBB3, TOP2A, PTEN genes’mRNA before the neoadjuvant chemotherapy.Result1. The expression of BRCA1, TYMS, TOP2A genes’mRNA were no relative to age, menstrual condition, tumor histological grade, metastasis of axillary nodes or expression of ER, PR, HER2or Ki-67;The expression of TUBB3gene’mRNA was relative to tumor histological grade(P=0.037), its high level was more common in the third grade of a tumor histological grade,it was no relative to age, menstrual condition, metastasis of axillary nodes or expression of ER, PgR, HER2or Ki-67;The expression of PTEN gene’mRNA was relative to ER/PR, its high level was more common in positive of ER/PR, it was no relative to age, menstrual condition, tumor histological grade, metastasis of axillary nodes or expression of HER2or Ki-67.2. The expression of BRCA1, TYMS, TUBB3, PTEN genes’ mRNA were no relative to molecular subclasses; the expression of TOP2A gene’mRNA was relative to molecular subclasses (P=0.021), its high level was more common in HER2+subtype.3. The expressions of TUBB3, TOP2A genes’mRNA were relative to the response of anthracyclines with taxanes (P=0.008, P=0.003), low TUBB3and high TOP2A were good response to anthracyclines with taxanes.4. A significant correlation with neoadjuvant chemotherapy efficiency and axillary node metastasis, expressions of ER, HER2, Ki-67, TUBB3was found. Age, menstrual condition, tumor histological grade, and expression of PR, BRCA1, TYMS, TOP2A, PTEN were not correlation with the response to treatment (P<0.05). And ER were regarded as protective factors of breast cancer (b>0).Conclusion1. The high expression of TUBB3gene’mRNA was more common in the third grade of a tumor histological grade.2. The high expression of PTEN gene’mRNA was more common in positive of ER/PR.3. The high expression of TOP2A gene’mRNA was common in HER2+subtype.4. The low expression of TUBB3and high expression of TOP2A genes’mRNA maybe have good response to anthracyclines with taxanes. |
