Objective: to separate SP cells from SGC-7901and identify theirbiocharacteristics.Methods: SGC-7901cells were sorted as SP cells and nonSP cells usingflow cytometry after SGC-7901cells were stained with Hoechst33342.theproliferation of two group cells in vitro was detected using CCK-8; the selfrenew ability was observed in serum-free culture media; the proportion ofSP cells was detected again after regular culture for a week; drug resistanceexperiment was used to observe two group cell’s resistance to5-FU;western-blot was performed to examine the protein expression of ABCG2and Bcl-2;the mRNA expression of Musashi-1and CD44was detected byreal-time PCR;the tumorigeness in nude mouses was observed after nudemouses had been subcutaneously injected SP cells and nonSP cells for8weeks.Results: The proportion of SP cells in SGC-7901is2.3+0.78%;compared with nonSP cells,SP cells have stronger proliferation ability andhigher resistance to5-FU;SP cells could form obvious tumor spheres and possess stronger asymmetric fission ability;in addition,SP cells have ahigher expresssion of Musashi-1and CD44at mRNA level and also have ahigher expression of ABCG2and Bcl-2at protein level; xenograftexperiment demonstrates that2×10~3SP cells could form tumor afterinjecting into nude mouse subcutaneously while2×10~4nonSP cells couldnot form tunor.Conclusion: There is a small proportion of SP cells in SGC-7901,whichhave relative biocharacteristics of tumor stem cells.
