Cell scratch assay lentiviral vector effects on SW480 cell migration. Cell adhesion assay lentiviral vector transfected SW480 cell adhesion ability between control. Invasion assay lentiviral vector changes SW480 cell invasion capacity.Results Mutant virus transfected SW480 cells compared with the wild type, mutant group expressed CO-029 abundantly, so that the amount of the protein molecules which form the protein networks has changed. Mutant virus transfected SW480 cells compared with the control group, it scratches heal quickly, indicating that the migration ability increase. This change has become increasingly apparent as time changes. The number of cells which adherent to each other is bigger in the control group, this reflect the adhesion ability decrease in the mutant group. Compared with control, the number of cells which move to the lower part of transwell is much bigger in the mutant group.Conclusion Tetraspanin-8 mutant lentiviral vector can change the expression of SW480 cells, which reflect the change of its molecular network functions; while increasing the migration and invasion capabilities of SW480 cell, its adhesion is also decreased, this all promote the metasis of tumor cell,CO-029 shows a significant effect.Part II:Effect of Niclosamide on the Growth and Apoptosis of Human Colon Cancer Cell LineObjective To investigate the inhibitory effect of niclosamide on the growth and cell cycle of human colon cancer cell line SW480, and to explore its possible mechanism.Methods CCK-8 was performed to reveal the inhibition of niclosamide on the growth of SW480. Flow cytometry was used to determine the change of cell cycle and apoptosis after treated with niclosamide. Western Blot assay was used to test the regulation of cell protein levels.Results Niclosamide can inhibit the growth of SW480 cell, its half inhibitory concentration is 4.6 ? mol/L. Niclosamide increased the percent of G0/G1 phase and decrease the percent of S phase in the cell cycle, and increase the cell apoptosis. Compared to the control, Nicosamide decrease the expression of (3-Catenin by 19.73%.Conclusion Niclosamide inhibit the growth of colon cancer SW480 cells, block its cell cycle in G0/G1 phase, induct the apoptos, inhibit the Wnt/?-Catenin signal pathway, niclosamide is a potential anti-cancer drug.
