Objective:To explore the effects and mechanisms of Qishen yiqi Decoction on the left ventricle hypertrophy and Myocardial fibrosis in mice with Ang II-induced hypertension.Methods:we used wild-type mice infused a dose of Ang II as research subjects of hypertension.Fifty male C57 mice, were randomly divided into five group: control group,Ang II group, Qishen yiqi decoction large-dose group(QSH), Qishen yiqi decoction small-dose group(QSL) and Captopril group( Cap). After 8 weeks treatment,systolic blood pressure(SBP) and Echocardiography were measured.pathologic changes of left ventricular myocardium were viewed with HE staining methed.CVF were detected by Van Gieson and SOD?MDA were detected by ELISA.In addition, the level of TNF-? and IL-6were measured by ELISA.the m RNA expression of ANP?BNP??-MHC were quantified by RT-PCR and the TLR4, I?B? and p65 protein were quantified by Western blot.Results:1. blood pressure in each of Ang II group were more than 150 mmhg,compared with control group,blood pressure in each of Ang II group were significantly higher. the model of Cardiac hypertrophy in mice was stable and successful. QSH?QSL?Cap group can had significantly declined LVEDd?LVESd values and increased EF?FS values compared with the model group.These are both better than QSH group with captopril group.2. Compared with the model group, heart weight and HMI decrease significantly and ANP?BNP??-MHC m RNA was decreased in the QSH group and Cap groups mice, These are both better than QSL group. 3. HE staining of left ventricular muscle in the model group, Cardiac muscle fiber arrayed disorderly. Drug intervention groups relieve cardiac muscle fiber arrayed disorderly and QSH group is better than other groups.Masson coloring:the model group could promate collagen surrounding blood vessel and QSH group?Cap group could also ameliorate collagen surrounding blood vessel in heart histologically. QSH group?QSL group and Cap groups could reduce HYP of of left ventricular in comparison with model group.4. The expression of proinflammatory factor IL-1 beta, trnnor necrosis factor, IL-6 could be reduced by Qishen yiqi decoction and Captopril groups curcumin compared with model group. 5.SOD activity of drug intervention groups increased and MDA content of drug intervention groups decreased in comparison with model group.6.Immunocytochemical staining analysis of NF-KB: model group:expression of NF-KB was enhance in cadiocyte endochylema, distributed wide spreadly in cadiocyte endochylema,distributed like trabs staining deeply. Drug intervention groups compared with model group, expression of NF-KB were decreased, amounts of fine particle in cadiocyte endochylema, diffused distribution. 7.The expression of TLR4/NF-k B signaling pathway:Phosphorylation and degradation of I?B? and phosphorylation of NF-k B p65 were significantly suppressed in Qishen yiqi decoction and Cap group. QSL group and Cap group inhibite TLR4/NF-k B signaling pathway activation campared with in model group.Conclusions: 1.In this study,the model of ventricular hypertrophy in mice with Ang II-induced hypertension was stable and successful. 2. Qishen yiqi Decoction can limit the development of left ventricular hypertrophy and Myocardial fibrosis induced by Ang II-induced hypertension in mice. The same effect to Captopril. 3. Qishen yiqi Decoction can limit the development of left ventricular hypertrophy caused by Ang II,which might through inhibition signal transdution of TLR4/NF-?B...
