and fed with high-sugar and high-fat diet to construct a diabetic cardiomyopathy model.By observing the general vital signs,weight,and blood glucose of rats,the rat’s model formation was judged.A rat model of type II diabetes was established,and high-sugar and high-fat feed was continued to build a model of diabetic cardiomyopathy.In order to determine the efficacy of the drug,the experiment was divided into five groups,namely the normal group,the model group,the positive control group,the Shenmai low-dose group,and the Shenmai high-dose group.Detect fasting blood glucose in rats,determine the hypoglycemic effect of the drug,calculate the cardiac index of rats,make cardiac pathological sections for observation,detect the ultrastructure of myocardium and autophagosomes,and determine the morphological changes of myocardium in rats.Heart damage was judged by serum creatine kinase(CK)andα-hydroxybutyrate dehydrogenase(HBDH)contents.Based on the levels of triglyceride(TG),aspartate aminotransferase(AST),and aspartate / alanine aminotransferase(AST/ALT)in the blood,determine whether the drug is reducing lipids and preventing fatty liver Aspect effect.2.Protective effect of Shenmai Fang on HUVECs endothelial cell cobalt chloride injury: establish a HUVECs endothelial cell cobalt chloride injury model,observe the cell growth status with an inverted fluorescence Shenmaifang high-dose group can significantly improve the above phenomenon,and the Shenmaifang group is more effective than metformin The improvement was obvious in the group and the lower dose group in the Shenmaifang high-dose group;The body weight of type II diabetic rats decreased significantly(P<0.001).The metformin hydrochloride group and Shenmaifang high-dose group could increase the weight of diabetic rats.The Shenmaifang low-dose group showed weight gain in diabetic rats two weeks before administration.;Fasting blood glucose and fasting blood glucose in the tail vein of type II diabetic rats increased significantly(P<0.001).Metformin hydrochloride group,Shenmaifang low-dose group,and Shenmaifang high-dose group did not significantly improve blood glucose in diabetic rats;II trend.The serum AST of the rats in the group had a downward trend.The Shenmaifang group had a significant decline compared with the metformin hydrochloride group,and the Shenmaifang low-and high-dose groups had a significant decline compared with the Shenmaifang low-dose group.The serum aspartic acid / alanine in type II diabetic rats Acid aminotransferase(AST/ALT)has an increasing trend.The serum significance(P<0.05);pathological observation of Shenmaifang could improve myocardial hypertrophy,necrosis,and fibrosis.,Has a protective effect on the heart;Shenmai formula can reduce the rate of apoptosis.2.Shenmai Fang’s protective effect on HUVECs endothelial cell cobalt chloride injury: Cobalt chloride injury can significantly reduce HUVECs endothelial cell survival rate(P <0.001),and Shenmai two groups can improve HUVECs endothelial cell survival rate(P<0.05).By fluorescence staining,it was found that compared with the normal group,the apoptosis of the model group was severe,the apoptosis of each administration group was reduced,the mitochondrial membrane potential of the model group cells was reduced,and the state of the Shenmai group was alleviated.Conclusion: 1.Shenmai Fang can improve the body weight,blood lipid,cardiac index,myocardial enzyme spectrum and liver function of diabetic rats,but it has no significant effect on blood glucose.Pathological sections showed that Shenmai Fang could effectively improve the degree of myocardial hypertrophy and fibrosis in diabetic rats,and had a protective effect on the heart.2.Shenmai Fang can promote cell proliferation,and has certain protective ability against hypoxic-damaged cells,which can reduce the apoptosis level of cells under hypoxic conditions,and the Shenmai injection group is more effective
