ObjectiveAfter the success of the establishment and identification of rabbit model of SCII,invigorate the circulation of intervention,having look after using tissue edema of SCII and the effect of Ca2+ concentration,and the spinal cord tissue Cytc,Hsp70,Bcl-2,the influence of Bax,Caspase-3,and compare TaoHong SiWu Decoction,invigorate the circulation of HuoXue YangXue Decoction prevention Cytc SCII model Caspase-3 the mechanism of action of mediated apoptosis pathway.Methods1.The studies of SC? animal modelBy blocking rabbit lumbar artery reperfusion after 30 min 3-6 1 h method to establish model of SCII,and through the digital subtraction angiography technique to observe blood supply situation,in order to make clear before ischemia,artery blocked blood flow recovery after ischemia and reperfusion,pathological observation of neurons in the spinal cord tissue damage situation,to identify the feasibility of this kind of building method.2.The influence of HuoXue YangXue Decoction on rabbit SCII model edema and Ca2+.HuoXue YangXue Decoction intervention in the rabbit model of SCII,TaoHong SiWu Decoction,observed by the method of calculation of spinal cord tissue water content qi and nourishing blood soup influence on the rabbit model of SCII edema,through semi-automatic biochemical analyzer is used to test the spinal cord tissue concentrations of Ca2+ discusses activating blood nourishing blood soup on the rabbit model of SC? Ca2+ concentration.3.HuoXue YangXue Decoction on rabbit model of SC? cytc caspase-3 mediated apoptosis pathway.HuoXue YangXue Decoction intervention in the rabbit model of SC?,TaoHong SiWu Decoction,by immunohistochemistry and enzyme-linked immune and rt-pcr methods Cytc,HSP70,Bcl-2,Bax,Caspase-3,invigorate the circulation of nourishing blood soup on the rabbit model of SC? Cytc-Caspase-3-mediated apoptosis pathway.Results1.Technology Digital subtraction angiography?dsa?observation block lumbar arteries,contrast agent to block the parts can't through the clip part later,when open the vascular clamp again,the contrast agent could move into blood vessels that was blocked again.HE staining,blank group of spinal cord neurons basic no abnormalities,pure ischemia group see cavity tissues surrounding neurons form,the nucleus position off center,indistinct nucleoli.Neurons in the spinal cord ischemia reperfusion group change spindle,the nucleus pycnosis,nucleoli disappear.See his surname dyeing,blank group than he's small body,form a complete,dyeing is blue,the nucleus round,nucleolus clear;Nissl body pure ischemia group was obviously reduced,neurons swelling,outline is not clear,the nucleus pycnosis,ischemia reperfusion group see nissl body is reduced,a large number of dissolving,edema occur around the nucleus pycnosis,indistinct nucleoli.Neurons,counting result is pure ischemia group and ischemia reperfusion group,compared to a blank neuron Numbers are down,there is obvious difference?p<0.01?,ischemia reperfusion group is purer group compared with significant difference?p<0.05?.2.Moisture testing see HuoXue YangXue Decoction and TaoHong SiWu Decoction group spinal cord water content decreased,compared with normal saline group had significant difference?p<0.05?,HuoXue YangXue Decoction is TaoHong SiWu Decoction group,with significant difference?p<0.05?.Ca2+ content detection see peach siwu decoction group and invigorate the circulation of HuoXue YangXue Decoction intracellular Ca2+ in the saline group,with significant difference?p<0.05?,HuoXue YangXue Decoction group of intracellular Ca2+ the TaoHong SiWu Decoction group,with significant difference?p<0.05?.3.The detection after drug intervention Cytc,HSP70,Bcl-2,the change of Bax,immunohistochemical results,TaoHong SiWu Decoction,invigorate the circulation of blood group compared with normal saline group,has significant difference?p<0.01?,HuoXue YangXue Decoction and TaoHong SiWu Decoction group comparison,Cytc there was no significant difference?p>0.05?,more than all have significant difference?p<0.05?.Enzyme-linked immune result,TaoHong SiWu Decoction soup,invigorate the circulation of blood group compared with normal saline group,has significant difference?p<0.01,p<0.05?,HuoXue YangXue Decoction and TaoHong SiWu Decoction,had significant difference?p<0.05?.Rt-pcr to detect the change of Caspase-3,TaoHong SiWu Decoction,invigorate the circulation of blood group compared with normal saline group,invigorate the circulation of HuoXue YangXue Decoction soup and TaoHong SiWu Decoction group,had significant difference?p<0.05?.Conclusions1.The method of SCII animal model comply with the requirement of experiment research.2.HuoXue YangXue Decoction can reduce the rabbit model of SCII oedema,can improve the rabbit model of SCII Ca2+ overload,so as to achieve control of SCII,and the effect is better than TaoHong SiWu Decoction.3.HuoXue YangXue Decoction can reduce Cytc SCII model,the expression of Bax,Caspase-3,increase the expression of HSP70 and Bcl-2,thereby inhibiting Cytc-from whole Caspase-3 mediated apoptosis,reach the role of prevention and treatment of SCII,and the effect is better than that of TaoHong SiWu Decoction.
