Objective:To detect the pathological changes of spinal cord tissue of rabbits and the expression of Nuclear factor-kappaB,vascular cell adhesion molecule-1 and cell adhesion molecule-1 during spinal cord ischemic-reperfusion injury via Huoxue Tongdu Decoction in order for its involved mechanism.Methods:According to the random number table,72 New Zealand rabbits which were 1.8-2.2kg and 3months old were randomly divided into three group:Sham-operated(n=8), Physiological saline(n=24), Huoxue Tongdu Decoction(n=24). Sham-operated group were fed normally. Huoxue Tongdu Decoction group were lavaged with Huoxue Tongdu Decoction for 7 days.Physiological saline group were lavaged with physiological saline for 7 days. Lumbar artery occlusion was performed along the third to sixth Lumbar artery root to induce spinal cord ischemia in Physiological saline group and Huoxue Tongdu Decoction group. Lumbar artery occlusion was not performed in the Sham-operated group. To observe the pathologic changes in spinal cord tissue, and detect the expression of NF-κB, VCAM-1 and ICAM-1 with Immunohistochemistry and ELISA at 0.5h, 1h,4h and 8h after reperfusion.Results:1. Pathological changes of spinal cord tissue:The nerve cells of Spinal cord were normal in Sham-operated group. There are many degenerative and necrotic nerve cells of Spinal cord in Physiological saline group. There are some swollen and necrotic nerve cells of Spinal cord in Huoxue Tongdu Decoction group.2. The result of Immunohistochemistry:The expression of NF-κB in Sham-operated group was lowest(P<0.01),and it was significantly higher in Physiological saline group than that of Huoxue Tongdu Decoction group at each time point(P<0.01, P<0.05).There were no VCAM-1 and ICAM-1 postive vessels in Sham-operated group.It appeared VCAM-1 positive blood vessels in Physiological saline group and Huoxue Tongdu Decoction group at 1h, and the former is higher than the latter at 1h,4h and 8h(P<0.01, P<0.05).It appeared ICAM-1 positive blood vessels in Physiological saline group and Huoxue Tongdu Decoction group at 4h, and the former is higher than the latter at 4h and 8h(P<0.01, P<0.05).3. The result of ELISA:The expression of NF-κB, VCAM-1 and ICAM-1 in Sham-operated group were low at each time point (P<0.01).The expression of NF-κB, VCAM-1 and ICAM-1 in Physiological saline group were highest,and they were significantly higher than that of Huoxue Tongdu Decoction group (P<0.01, P<0.05).Conclusions:1. Huoxue Tongdu Decoction can reduce the necrosis of nerve cells in ischemic region to improve spinal cord ischemia reperfusion injury.2. Huoxue Tongdu Decoction can inhibit the activation of Nuclear factor-kappa B, and delay the action between, vascular cell adhesion molecule-1 and cell adhesion molecule-1.So it can reduce the infiltration and aggregation of leukocytes, and reduce the inflammation and the necrosis of nerve cells in the early reperfusion.
