Purpose:1.To explore the relationship between TCM constitution,age and perceived stress in healthy population.2.To analyze the similarities and differences between the gray matter volume and function connection of perceived stress in different age groups,and to explore the changes of the structure and function of mental stress related brain area during the brain development.3.To analyze the correlation between the gray matter volume and functional connectivity index and cognitive function in different age groups,and to explore the possible mechanism of perceived stress's effect on cognitive function.Method:1.A total of 120 healthy subjects including adolescents,young people and adult were recruited to attend this study.The TCM Constitution scale was used to measure the TCM constitution and perceived stress scale(PSS)was used to measure the perceived stress level.A single factor analysis of variance(ANOVA)or multiple independent sample nonparametric tests was used to compare the PSS scores of different groups,the PSS scores difference of different TCM constitution and the PSS scores of different TCM constitution in different age groups.SPSS23.0 software was used to analyze the data.When the P value was less than or equal to 0.05,the difference was statistically significant.2.120 participants(adolescence,young adult,adult)who were eligible for inclusion criteria were included in the study.The perceived stress level was measured by pressure perception scale(PSS).The cognitive function,such as attention.working memory,risk decision ability,learning and memory,were measured by Stroop color test,N-back test,balloon simulation risk task(BART)and pairing learning test(PAL).The functional MRI was performed by 3.0T GE magnetic resonance,and the structures(T1)and the resting function of the subjectswere collected.The data were analyzed by SPM12 software based on Matlab2015b.Brain gray matter volume was measured by Voxel-based morphological(VBM)method.Resting-state functional connectivity(rsFC)and spectual dynamic causal model(sDCM)were used to observe the changes of brain function.The the peak intensity of the brain region was extracted and a correlation analysis between it and the test results of the cognitive function was analyzed.Results:1.There was a significant difference of PSS scores between the three groups(P<0.01).The scores of the PSS were higher in the adolescent group than in the young group and the adult group.There was nosignificant difference between the young group and the adult group(P>0.05).2.There.was significant difference of PSS scores difference between qiyu constitution and other TCM constitutions(P<0.05).3.Further explore the relation in in different age groups,results show that there was still a significant difference of PSS scores difference between qiyu constitution and other TCM constitutions(P<0.05).4.The scores of Stoop color,Stroop word,Stroop color word,N-back accuracy in three age groups were statistically different(P<0.05),but for Stroop anti-interference time,PAL correct rate and total number of PAL BART average inflated number,BART blasting balloon percentage,BART single balloon completion time in three age groups,there was no statistically significant difference(P>0.05).5.Analysis of the correlation between PSS and cognitive function in the three groups showed that the PSS score was significantly correlated with the percentage of BART blasting balloon(r = 0.52,P = 0.02)after control of IQ and sex in the youth group.There was no significant correlation between PSS score and cognitive function index(P>0.05).6.The difference of the correlation between gray matter volume and PSS score in different age groups was observed by VBM.The interaction between PSS and age on VBM results showed a significant cluster(P(2.66)>7.68,P<0.001)which the inculde amygdala,insula,temporal and parahippocampal gyrus.In the post hoc analysis,for each age group,there was a significant positive correlation between VBM and PSS in adolescents(r2 = 0.56,P<0.01).There was a negative correlation between VBM and PSS in young and adult groups(r2 = 0.33,P<0.01)(r2 = 0.22,P = 0.07).7.In the different age groups,after controlling the sex and IQ,there was a positive correlation between the volume of orbital frontal gray matter and the completion time of Stroop color(r = 0.49,P = 0.03).There was a positive correlation between the volume of orbital frontal gray matter and the anti-interference time of Stroop(r = 0.48,P =(R =-0.51,P = 0.02).There was a negative correlation between gray matter volume of insula and the rate of BART blasting balloon(r =-0.45,P = 0.04),there was a negative correlation between the orbital frontal gray and the rate of BART blasting balloon(r =-0.51,P =).There was a significant postive correlation between the temporal pole gray matter volume and the time of Stroop color word(r = 0.56,P = 0.01).No significant results among other index were found(P>0.05).8.Using right amygdala as seed,there was a significant interaction effect rate of PSS and age in rsFC showed that the interaction of PSS appeared in the medial prefrontal cortex(vmPFC)and corrected P =0.04.Further post hoc analysis showed that rsFC(T test)in the vmPFC had a significant difference between the adolescent and adolescent groups(P<0.001).No significant differences in rsFC were observedbetween young adults and adults.In adolescents,there was a positive correlation between the PSS and amygdala-vmPFC rsFC(r2 = 0.56,P<0.001).In the young group,there was a negative correlation between the PSS and amygdala-vmPFC rsFC(r2 = 0.28,P = 0.01).There was no significant interaction between age and PSS in rsFC when left amygdala as seed.9.When control for IQ and gender,in young adult group,there was a negative correlation between thersFC of amygdala-vmPFC and the percentage of BART blasting balloon(r =-0.54,p = 0.01)and no significant correlation was found in the other group(P>0.05).10.Spectrum dynamic causal model analysis shows that among young people,the winning model is an effective connection from the amygdala to vmPFC with a probability=1 and a connection strength of 0.067 Hz.In the youth group,the winning model is from vmPFC to the amygdala probability of 0.96,the connection strength of 0.068.Conclusion:1.For different age groups,the level of psychological stress may be different.In the adolescent group,the perceived stress may be higher than that of the youth group and the adult group.2.TCM constitutions,Qiyu constitution group's perceived stress may be higher than other constitutions.3.The different levels of stress perception in different age groups may be related to the structural changes of brain pressure-related areas,especially in the areas of amygdala,insula,obrital frontal cortex,tempral pole and parahippcampus.4.The amygdala-vmPFCrsFC is associated with stress,and there is a functional direction change in the process of developing from adolescents to young people.5.The maturation of the amygdala-vmPFC loop is related to the risk-taking behavior,and the vmPFC-amygdala loop may regulate the risk-taking behavior.
